摘　　要：背景与目的:肿瘤原位移植模型已成为肿瘤实验动物学的主要载体,但目前尚缺乏方便可靠的方法监测模型体内肿瘤的生长变化。本文评价β-人绒毛膜促性腺激素(βhumanchorionicgonadotropin,β-HCG)系统对肿瘤原位模型内肿瘤生长的监测作用。方法:将pclone-β-HCG质粒稳定转染的人卵巢癌细胞系A2780-CG制成裸小鼠原位移植模型,通过连续检测鼠尿中β-HCG/肌酐值,绘制曲线,间接了解小鼠体内肿瘤生长规律,以及顺铂腹腔化疗后,该比值对顺铂抗瘤效果的监测作用。结果:β-HCG系统转染瘤细胞A2780-CG的成瘤时间和母代相同。小鼠尿中β-HCG/肌肝值与体内瘤重量呈正相关(r=0.98);顺铂腹腔化疗后,β-HCG/肌酐值呈进行性下降。结论:β-HCG系统可无创、敏感地监测卵巢癌原位移植模型中肿瘤生长规律,为该模型在肿瘤治疗中的应用提供简便的监测手段。Background &Objectives:Tumor orthotopic transplantation model has become the main carrier for t umor animal experiment.However,there w as no convenient and reliable method to monitor tumor growth in animal bod y.This study was designed to evaluate the mo nitoring impact of urinaryβ-human chorionic gonadotropin(β-HCG)on tumor growth in orthotopic implantation model.Methods:pclone-β-HCG stably transfected human ovarian carcinoma cell line A2780-CG was made as the orthotopic implantation model.The urinaryβ-HCG /creatine ratio was continuously determined and plotted into curve,so as to indirectly know t he rule of tumor growth in nude mice an d the monitoring value of the ratio fo r intraperitoneal chemotherapy by cisplatin.Results:The tumorigenesis time inβ-HCG system transfected tumor cell A2780-CG was similar to that in the original cell line.The content of urin aryβ-HCG /creatine had a positive correlation to tumor weight in animal body(r=0.98);After chemotherapy with cisplatin,theβ-HCG /creatine ratio showed progressively decreased.Conclusion:β-HCG system could be used to sensitiv ely and non-invasively monitor the t umor growth in orthotopic implantation model of hu man ovarian carcinoma in nude mice.Meanwhile,it provides a new co nvenient monitoring method for tumor treatment in the mod el.
