摘  要:目的:研究人参皂苷Rd(ginsenoside Rd,GRd)的镇痛作用及对小鼠中枢蛋白激酶γ(proteinkinase Cγ,PKCγ)表达的影响。方法:采用扭体和福尔马林舔足法观察GRd的镇痛作用,侧脑室注射CaCl_2和维拉帕米(Verapami l,Ver)后,通过扭体法分析GRd的镇痛作用及其与Ca^(2+)的关系;免疫组织化学方法检测GRd对小鼠脊髓背角、大脑皮层和丘脑PKCγ免疫阳性细胞表达的影响。结果:GRd对冰醋酸所致的扭体及福尔马林所致的舔足反应均有抑制作用(P<0.05);给予CaCl_2和Ver后,GRd的镇痛作用被CaCl_2拮抗,被Ver加强(P<0.01);GRd可明显减少小鼠脊髓背角、大脑皮层和丘脑PKCγ免疫阳性细胞(P<0.01)。结论:GRd的镇痛作用涉及外周和中枢,Ca^(2+)与PKCγ参与了GRd的镇痛作用。Objective: To study the analgesic effects of ginsenoside Rd(GRd) and its influence on the expressions of central protein kinase Cγ(PKCγ). Methods: Analgesic effects of GRd were observed by using writhing and formalin licking foot method, analgesic effects of GRd and its connection to Ca^(2+) were analyzed by using writhing method after intracerebroventricular injection of CaCl_2 and Verapamil(Ver), the influence of GRd on the expressions of immunoreactive cells PKCγ incornu dorsale medullae spinalis, pallium and cerebral ganglia of the mice were detected by immunohistochemical method. Results: GRd showed inhibitory effects on glacial acetic acid-induced writhing method and formalin licking foot method(P〈0.05); after injecting CaCl_2 and Ver, analgesic effects of GRd were attenuated by CaCl_2 while enhanced by Ver(P〈0.01); GRd could obviously reduce the expressions of immunoreactive cells PKCγ incornu dorsale medullae spinalis, pallium and cerebral ganglia of the mice(P〈0.01).Conclusion:Analgesic effects of GRd are related to peripheral nerve and central nerve, simultaneously Ca^2+ and PKCγ have participated in analgesic effects of GRd.
