摘 要:目的探究LKB1基因对胰腺癌细胞上皮间质转化及侵袭迁移的影响情况。方法通过细胞实验检测LKB1过表达对胰腺癌上皮间质转化相关蛋白E-钙黏蛋白,及间质标记物N-钙黏蛋白、vimentin的影响,并分析其对人胰腺癌细胞PANC-1迁移和侵袭能力的影响。通过免疫荧光法对转染后的细胞进行处理,并通过荧光显微镜高倍镜观察细胞的变化情况。结果与空载体组、转染试剂组相比,LKB1过表达组中LKB1蛋白及E-钙黏蛋白表达水平显著升高(P<0.05);间质细胞标志物N-钙黏蛋白、Vimentin的蛋白相对表达水平明显降低(P<0.05)。LKB1过表达对胰腺癌细胞PANC-1迁移和侵袭能力的影响分别为转染试剂组(45.56±6.60)、空载体组(45.06±3.85)、LKB1过表达组(22.54±3.74);转染试剂组(111.48±14.90)、空载体组(107.58±13.18)、LKB1过表达组(57.48±7.94);LKB1过表达组细胞迁移和侵袭能力均显著降低(t=-14.501、-14.824、-12.810、-13.808,P均=0.000)。结论 LKB1可以抑制人胰腺癌细胞的迁移和侵袭,该抑制过程可能与其参与上皮间质转化有关。Objective To investigate the effect of LKB 1 gene on epithelial mesenchymal transition, invasion and migration of pancreatic cancer cells. Methods Through the proliferation test of LKBI overexpression on pancreatic cancer epithelial mesenchymal transition related protein E-cadherin and mesenchymal markers of N- cadherin and vimentin, and analyzes its influence on the migration and invasion of human pancreatic cancer cell line PANC-1. After transfection, the cells were treated with immunofluorescence, and the changes of cells were observed by fluorescence microscopy. Results After human pancreatic cancer cells were transfected with plasmid over-expressed LKB1, protein expression level of LKB 1 was significantly enhanced compared to the transfection agent group; the ability of cell invasion and migration was remarkbly decreased (P〈0.05); The effects of LKB1 overexpression on the migration and invasion ability of PANC-1 in pancreatic cancer cells were respectively the transfection reagent group (45.56±6.60), the empty vector group (45.06±3.85), and the LKB1 overexpression group (22.54±3.74). The reagent group (111.48±14.90), the empty carrier group (107.58± 13.18), LKB 1 overexpression group (57.48±7.94); Level of related mesenchymal protein was distinctly reduced (t=-14.501, -14.824, -12.810, -13.808, all P=0.000). Conclusion LKB1 can inhibite the invasion and migration of human pancreatic cancer cells, and this process also probably is related to the epithelial mesenchymal transition.
