摘  要:目的 探讨P-选择素(P-selectin)在大鼠全脑缺血再灌注血管内皮细胞损伤过程中的作用。方法将大鼠随机分为假手术组和脑缺血30min再灌注1h、3h、6h、12h、24h、48h、72h组,动物模型采用全脑缺血模型三血管阻塞法,用免疫组化法检测脑组织缺血再灌注不同时间点FⅧ相关抗原(vWF)的表达以标记内皮细胞损伤与修复过程,同时用原位杂交方法检测P-选择素mRNA的表达变化。结果 假手术对照组未见P-选择素mRNA表达,脑缺血再灌注后1h表达出现上调,12~24h达高峰,72h仍有表达。FⅧ相关抗原在假手术组呈强阳性表达,脑缺血再灌注后3h表达下降,24~48h表达最低,72h表达开始恢复。结论 脑缺血再灌注微血管内皮细胞损伤主要发生在早期,此过程中P-选择素mRNA表达上调,说明P-选择素参与了脑缺血后内皮细胞的损伤过程。Objective To study the effect of P-selectin in inflammatory damage of endothelial cell in global cerebral ischemia-reperfusion. Methods The rats were randomly divided into the sham operated control group and lh,3h,6h,12h,24h,48h,72h reperfusion group. The global crebral ischemia-reperfusion model was performed by occluding three arteries. The expression of vWF was evaluated by immunohistochemical method and P-selectin mRNA was detected by in situ hybridization. Results P-selectin mRNA was not detected in sham operated con-trol group. It was detected as early as 1h after reperfusion and peaked at 12-24h after reperfusion. At 72h after reperfusion. It still could be detected. The expression of vWF was strong positive in sham operated control group. At 3h after reperfusion ,the expression of vWF began to decrease and its lowest level appeared during 24~48h. At 72h, the expression began to regain. Conclusion The damage of microvascular endothelial cells appeares in early stage after cerebral ischemia-reperfusion. P-selectin is involved in the inflammatory damage of endothelial cell dur-ing global cerebral ischmia-reperfusion.
