摘  要:目的 研究可溶性白细胞介素-2受体(sIL-2R)与肿瘤坏死因子-α(TNF-α)在实验性变态反应性脑脊髓炎(EAE)免疫学发病机制中的变化与作用。方法 应用豚鼠诱导EAE动物模型。在髓鞘碱性蛋白(MBP)与弗氏完全佐剂(CFA)免疫豚鼠的第8、15、22天处死动物,取脾细胞加笔入ConA诱生培养,收集上清液待测。采用酶联免疫吸附试验(ELISA)方法测定sIL-2R水平,采用生物活性测定法检测TNF-α水平。结果 EAE组的sIL-2R与TNF-α水平显著高于正常对照组。结论 sIL-2R与TNF-α在EAE兔疫学发病机制中具有重要作用。本实验为深入研究多发性硬化(MS)的发病机制提供了理论依据,给MS免疫特异性治疗寻觅到新的线索。Objective To investigate the changes and effects of sIL-2R and TNF-αon the im-munopathogenesis of experimental allergic encephalomyelitis(EAE). Methods EAE were induced in guinea pigs. The EAE were killed on 8th, 15th and 22nd day after immunization with myelin basic protein (MBP) and complete freund adjuvant (CFA). The spleen cells were collected and cultured with addition of ConA. And then the super-natant was obtained. The level of sIL-2R was examined by ELISA and the level of TNF-αwas detected by bioas-say. Results The levels of sIL-2R and TNF-αin EAE group were obviously higher than the control. Conclusion sIL-2R and TNF-αplay an important role in the immunopathogenesis of EAE. This experiment has offered theo-retical evidence for the further study of the pathogenesis of multiple sclerosis (MS) and finds new clues for the spe-cific immunologic treatment of MS.
