摘 要:目的:探讨芹菜素对人乳腺癌MDA-MB-231细胞的凋亡作用及相关机制。方法:常规培养人乳腺癌MDA-MB-231细胞,取对数期生长细胞分为芹菜素40、80、160μmol/L组和空白对照组,分别用芹菜素40、80、160μmol/L和生理盐水进行处理。处理24、48h后,采用MTT法检测各组细胞的增殖情况,流式细胞仪检测处理48h后凋亡和细胞周期情况,Western blot方法检测处理48h后,Pro-Caspase-3、Caspase-3和Caspase-6蛋白的表达情况。结果:MTT和流式结果显示,芹菜素80、160μmol/L处理组在24、48h均可显著抑制MDA-MB-231细胞的生长,且160μmol/L处理组比80μmol/L处理组的抑制作用更强。40μmol/L处理组在24h显示对MDAMB-231细胞的生长有一定作用,但与对照组相比无统计学差异。Western blot结果显示,芹菜素40、80、160μmol/L组和空白对照组处理细胞48h后,Caspase-3和Caspase-6蛋白表达水平随着芹菜素浓度的增高而依次上调,同时Pro-Caspase-3蛋白表达逐次降低。结论:芹菜素对人乳腺癌MDA-MB-231细胞增殖有抑制作用,并能诱导细胞的凋亡,其作用机制可能与Caspase-3、6信号通路的激活有关。Objective:To study the apigenin induced apoptosis of human breast cancer cell line MDA-MB-231 and its mechanism.Methods:Conventional culture human breast cancer cell line MDA-MB-231.The cells in logarithmic phase were divided into 40,80,160μmol/L apigenin group and control group,and treated with 40,80,160μmol/L apigenin and saline respectively.After 24 and 48 htreatment,cell proliferation was measured using the MTT assay to detect every group.Flow cytometry was used to detect the rate of apoptotic and cell cycle for MDA-MB-231 cells after treated with 40,80,160μmol/L apigenin and saline for 48 h.The protein expression of Pro-Caspase-3,Caspase-3 and Caspase-6 were detected by Western blot in every group after 48 htreatment.Results:MTT and flow cytometry assay showed that both 80,160μmol/L apigenin treated group significantly inhibit the proliferation of MDA-MB-231 cells after 24,48 h,and the inhibition of 160μmol/L treated group was stronger than 80μM treated group.50μmol/L apigenin treated group had an inhibitive effect compared with the control group,but there was no significant difference.Western blot revealed that the expressions of Caspase-3 and Caspase-6 were up-regulated with the increased of apigenin concentration,as well as the expression of Pro-Caspase-3 was decreased.Conclusion:Apigenin exhibits an inhibitory effect of the proliferation and promote apoptosis of MDA-MB-231 cells.The mechanism may be associated with the activation of Caspase-3,6 signaling pathway.
