摘  要:通过体外培养SH-SY5Y细胞,Tunel法观察五味子甲素(Schisandra A,Sch A)对MPP+染毒SH-SY5Y细胞形态的影响;激光共聚焦显微镜观察各组细胞核的变化;Western-Blot检测各组SH-SY5Y细胞中ERK及p-ERK蛋白表达水平的不同,探讨Sch A对1-甲基-4-苯基吡啶离子(1-methyl-4-phenylpyridinium ion,MPP+)引起的人神经母细胞瘤SH-SY5Y细胞凋亡及ERK信号转导通路的影响。结果发现,与正常对照组相比,1mmol/L MPP+染毒可引起SH-SY5Y细胞形态明显改变,凋亡细胞明显增多;各浓度组Sch A干预均可有效抑制SH-SY5Y细胞的凋亡。与对照组比较,MPP+组细胞中p-ERK蛋白表达明显抑制;而与MPP+染毒组相比,Sch A干预组p-ERK蛋白表达明显升高(P<0.05),且呈剂量依赖效应。结果表明,MPP+具有促进SH-SY5Y细胞凋亡的作用,而Sch A能明显缓解MPP+引起的SH-SY5Y细胞凋亡,该作用可能与ERK信号转导通路活性改变有关。To explore the pharmacological actions and molecular mechanisms of Sch A on the apoptosis of MPP^+to human neuro-blastoma SH-SY5 Y cells. The changes of morphological characteristics in SH-SY5 Y cells were analyzed by acridine orange(AO)/ethidium bromide(EB) staining and Tunel Kit. Subsequently,ERK and p-ERK levels were analyzed by western-blot assay. Com-pared with control group,a marked up-regulation of apoptosis was observed in SH-SY5 Y cells after being treated with 1mmol/L for48 h. Sch A intervention in each concentration group could effectively inhibit the apoptosis of SH-SY5 Y cells. It also showed thatthe expression of p-ERK protein in MPP^+group was significantly inhibited when compared with the negative control group, whilethe p-ERK level in each Sch A intervention group was significantly higher when compared with MPP+exposed group(P〈0. 05) Adose-dependent effect was observed. The data indicate that MPP^+has the effect on promoting apoptosis in SH-SY5 Y cells,andSch A can obviously alleviate the apoptosis of SH-SY5 Y induced by MPP^+,which may be related to the changes of ERK signal trans-duction pathways.
