摘 要:目的研究罗汉松实(花托)的急性毒性和亚慢性毒性,并对其安全性进行评价。方法急性毒性试验采用最大给药量法,以160.0 g/(kg·d)的剂量分2次小鼠灌胃200%罗汉松花托汁,观察14 d内的毒性反应。亚慢性毒性试验分3组,实验组分别以20 g/kg和10 g/kg的剂量,SD大鼠灌胃200%罗汉松花托汁,对照组给予蒸馏水,连续灌胃90 d。实验结束时采血测血液学和血生化指标并进行组织病理学检查,计算脏器系数。结果各组动物未见明显毒性反应,也未见死亡。实验组脏器系数、血液学和血生化指标与对照组相比,差异均无统计学意义(P>0.05)。组织病理学检查也未见与受试物相关的病理学改变。结论在本实验条件下,未见罗汉松花托汁对实验动物有明显的毒性反应,罗汉松花托安全性较高,可作为食品原料开发利用。Objective To study the acute and subchronic toxicity of Podocarpus macrophyllus fruit and evaluate its safety.Methods Maximum dosage method was adopted for acute toxicity experiment. The mice were administrated with the receptacle juice( with mass concentration of 200%) of Podocarpus macrophyllus fruit by intragastric infusion twice a day with total dosage of 160. 0 g/( kg·d). After being administrated,the mice were observed for 14 days. In subchronic toxicity test,rats were randomly divided into three groups. The rats of treatment groups were given the juice( with mass concentration of 200%) by oral gavage with dosage of 20 g/kg and 10 g/kg for 90 days continuously. And the rats of control group were given distilled water by oral gavage. The routine blood test,serum biochemical index determinations,and histopathological examination were carried out at the end of the experiment. The important organs were weighed to calculate the organ/body ratios. Results During the experiments,no toxic effects and death were observed in each group. The index determination and histopathological results showed no statistical significance on the difference between the treatment and control group( P〈0. 05). Histopathological findings did not correlate with histopathological changes in the subjects. Conclusion In this experiment,receptacle juice of Podocarpus macrophyllus fruit shows no acute and subchronic toxicity to animals. It can be used as raw materials for food development and utilization.
