摘　　要：目的探讨pERK蛋白在胃癌及癌旁组织中的表达及其与肿瘤恶性行为的相关关系。方法收集98例胃癌患者的胃癌组织及癌旁组织标本,应用免疫组化检测其中pERK蛋白表达情况,并根据胃癌组织pERK蛋白表达阳性情况将其分为pERK蛋白(+)组74例、pERK蛋白(-)组24例。采用荧光定量PCR法检测胃癌组织中增殖、侵袭、自噬基因mRNA表达量,评估pERK蛋白阳性表达对上述基因mRNA表达量的影响。结果胃癌组织中pERK蛋白阳性表达率显著高于癌旁组织,差异有统计学意义(P<0.05)。pERK蛋白(+)组增殖基因EZH2、PIK3CD、I2PP2A mRNA表达量高于pERK蛋白(-)组,WTX mRNA表达量低于pERK蛋白(-)组;侵袭基因E-cadherin、Syndecan-1 mRNA表达量低于pERK蛋白(-)组,HPA-1 mRNA表达量高于pERK蛋白(-)组;自噬基因Beclin1、MAP1LC3 mRNA表达量低于pERK蛋白(-)组,差异有统计学意义(P<0.05)。结论 pERK蛋白在胃癌组织中阳性表达率高,且存在pERK蛋白阳性表达的胃癌组织肿瘤恶性程度高。Objective To study the expression of phospho-extracellular signal-regul kinase（ pERK） protein in gastric cancer and adjacent tissues and its relationship with malignant behavior. Methods Gastric cancer tissues and paracancerous tissues were collected from 98 cases of gastric cancer patients,and immunohistochemistry was used to detect the expression of pERK protein; according to the positive pERK protein expression in gastric cancer tissues,all patients were divided into pERK protein（ ＋） group（ n = 74）,pERK protein（-） group（ n = 24）. The expression of proliferation,invasion and autophagy genes mRNA in gastric cancer tissue were detected by fluorescence quantitative PCR,and the influence of pERK protein positive expression on the expression of mRNA genes above were evacuated. Results The positive expression rate of pERK protein in gastric cancer tissues was significantly higher than that in adjacent tissues,with the difference statistically significant（ P〈0. 05）. The expressions of EZH2 PIK3 CD,I2PP2A genes mRNA in pERK protein（ ＋） group were higher than those in pERK（-） group,WTX mRNA expression was lower than that in pERK protein（-） group; E-cadherin,Syndecan-1 mRNA expression were lower than those in pERK（-） group,HPA-1 mRNA expression was higher than that in pERK protein（-） group; the expressions of MAP1LC3 and Beclin1 mRNA were lower than those in pERK protein（-） group,with the difference statistically significant（ P〈0. 05）. Conclusion The positive expression rate of pERK protein in gastric cancer tissue is high,and there is high degree of malignancy in gastric cancer tissues with pERK protein positive expression.
