摘  要:目的探讨淫羊藿苷对阿尔茨海默病(Alzheimer's disease,AD)大鼠学习记忆、海马区基质金属蛋白酶-9(MMP-9)、MMP-2及血脑屏障紧密连接蛋白ZO-1表达的影响。方法采用Aβ1-42海马注射建立AD大鼠模型,使用淫羊藿苷进行干预。以Morris水迷宫检测大鼠的空间学习记忆能力,采用Western-blot法分析紧密连接蛋白ZO-1的表达,用免疫组织化学法检测大鼠海马区MMP-9、MMP-2表达。结果与对照组相比,AD模型组大鼠学习记忆能力明显减退,ZO-1表达明显减少(P<0.05),MMP-9、MMP-2蛋白明显增加;淫羊藿苷干预后,大鼠学习记忆能力均明显提高,海马区MMP-9/MMP-2表达减少(P<0.05),ZO-1表达明显增加(P<0.05),以淫羊藿苷高剂量干予后明显。结论淫羊藿苷能通过抑制MMP-9/MMP-2表达,减轻血脑屏障破坏,从而改善AD症状。Objective To explore the effect of Icariin on learning and memory,Metalloproteinases(MMPs) expression and brain barrier permeability of hippocampus region in rats with Alzheimer's disease(AD).Methods Rats models of Alzheimer's disease were established by direct β-amyloid protein(Aβ1-42) injection method.For Icariin intervention test,the learning and memory ability was observed.The expressions of MMP-9/MMP-2 were analyzed by and Immunohistochemistry.The brain barrier permeability was detected by measured junction protein(ZO-1) expression.Results The learning and memory ability was obviously decreased in AD model rats.The expression of MMP-9/MMP-2 protein level were was higher compared to control group.After Icariin treatment.The the learning and memory ability was improved,and it could also inhibit the MMP-9/MMP-2 activity(P〈0.05) compared to AD model rats,the level of ZO-1 expression was significantly reduced.Conclusion Our result suggests that the protective role of Icariin in AD may be inhibit MMPs activity and maintaining BBB integrity could be another interesting therapeutic target at AD.
