摘 要:探讨一次跳绳运动对男大学生外周血单个核细胞骨代谢因子mRNA表达的影响。方法:7名骨密度、体脂正常的男大学生进行5分钟跳绳运动,运动前、运动后即刻、运动后恢复60分钟分别抽取静脉血,Ficoll密度梯度离心法分离单个核细胞,并提取外周血单个核细胞RNA,荧光定量PCR测定外周血单个核细胞IL-1β、IL-6、TNF-α、IFN-γ、OPG、RANKmRNA的表达。结果:与运动前相比,运动结束后即刻IL-1β、IFN-γ、OPG、RANK mRNA表达水平升高,IL-6、TNF-αmRNA表达水平无显著性差异;运动结束后60分钟OPG、RANKmRNA表达水平仍显著高于运动前。结论:一次无氧运动强度的跳绳运动暂时性提高了骨代谢水平,外周血单个核细胞IL-1β等因子从转录水平参与了运动调节骨代谢的过程,可作为反映骨代谢水平的分子标记。To investigate the effect of a single bout rope skipping exercise on mRNA expression of bone metabolism-related factors in peripheral blood mononuclear cell( PBMC) of male college students. Methods: 7 male college students with normal bone mass and body fat played rope skipping for 5 minutes. Before exercise,immediately after exercise and 60 minutes after exercise,venous blood was collected. PBMC was harvested,and the mRNA expression of IL-1β,IL-6,TNF-α,IFN-γ,OPG and RANK were detected using QRT-PCR. Results: Compared with the level before exercise,the levels of IL-1β,IFN-γ,OPG and RANK mRNA were increased immediately after exercise,and the expression of IL-6 and TNF-α mRNA was not significantly different from that before exercise. OPG,RANK mRNA expression level was still significantly higher than that before exercise.Conclusion: A single bout rope skipping exercise with anaerobic intensity temporarily improved the level of bone metabolism-related factors in PBMC,IL-1β and other factors in PBMC involved in the process of regulating bone metabolism from the transcription level,which could be used as molecular markers to reflect the level of bone metabolism.
