摘 要:目的研究AKT、VEGF和PTEN蛋白在脑动静脉畸形中的免疫组化表达,探讨PI3K/AKT/PTEN信号途径与脑动静脉畸形血管生成的关系。方法用免疫组化方法检测35例脑动静脉畸形组织和10例正常脑组织中PTEN、AKT(pan)、p-AKT(ser473)和VEGF蛋白的表达情况。结果 PTEN、AKT(pan)和VEGF在脑动静脉畸形血管内皮细胞中的免疫组化阳性表达率分别为80.00%、91.43%和80.00%,而在正常对照组阳性表达率分别为30.00%、40.00%和50.00%,差异均有统计学意义(均<0.05);而p-AKT(ser473)在脑动静脉畸形和正常对照组中的阳性表达率分别为11.43%和10.00%,差异无统计学意义(>0.05)。结论 PTEN的高表达可能在脑动静脉畸形的血管生成中起着一定的影响作用,它明显抑制了PI3K/AKT的信号途径,却仍旧表达了高水平的VEGF,推测PTEN不是通过PI3K/AKT的信号途径调控脑动静脉畸形的血管生成。Objective To investigate the expression and clinical significance of AKT, VEGF and PTEN proteins in the cerebral arteriovenous malformations(AVMs), and to discuss relationship between PI3 K/AKT/PTEN signal pathway and angiogenesis of AVMs. Methods The expressions of PTEN, AKT(pan), p-AKT(ser473) and VEGF proteins were examined by the Envision TM immunohistochemical staining in 35 specimens of AVMs and ten cases of normal brain tissues. Results The positive immunohistochemical expression rate of PTEN, AKT(pan) and VEGF in the AVMs were 80.00%, 91.43% and 80.00%,respectively. While, they were 30.00%, 40.00% and 50.00% respectively in the controlled group, and there was a significant difference between the two groups( < 0.05). Otherwise, the positive immunohistochemical expression rate of p-AKT(ser473) in the AVMs was only 11.43%, compared with the controlled positive expression rates of 10.00%; and there was no significant difference( > 0.05). Conclusions The abnormal high expression of PTEN in AVMs may play a certain role in the angiogenesis.There was still a high expression level of VEGF, although it significantly inhibited the PI3 K/AKT signaling pathway, suggesting that PTEN is not mediated by PI3 K/AKT pathway to regulate angiogenesis of AVMs.
