N-WASP在肾透明细胞癌组织及癌旁正常肾组织中的表达及与肿瘤病理分级和临床分期的相关性分析

摘　　要：目的探究神经-奥德里奇综合征蛋白(N-WASP)在肾透明细胞癌组织及癌旁正常肾组织中的表达及与肿瘤病理分级和临床分期的相关性。方法选取医院2012年6月-2014年6月期间收治的、符合纳入标准的肾癌患者70例。所有患者均行根治性肾切除术或肾部分切除术后,取标本制成肾透明细胞癌组织芯片及癌旁正常肾组织芯片,各70例,患者均随访3年;应用免疫组织化学染色,对比两种芯片之间N-WASP表达水平差异;并分析N-WASP表达水平与肿瘤病理分级和临床分期的相关性;对肾透明细胞癌患者生存率进行单、多因素分析。结果癌组织的N-WASP表达水平为(75.54±8.48)低于癌旁组织的(121.85±10.46),差异有统计学意义(P<0.05);随访3年后,N-WASP表达为低、中水平者生存率为80.95%高于高水平者的25.00%(P<0.05);年龄及肿瘤直径为肾透明细胞癌患者生存的相关因素(P<0.05);N-WASP表达水平为肾透明细胞癌患者生存的独立危险因素(P<0.05)。结论 N-WASP在肾透明细胞癌组织表达水平降低,为肾透明细胞癌患者生存的独立预测因素。OBJECTIVE To explore the expression of neural Wiskott-Aldrich syndrome protein（N-WASP）in clear cell renal cell carcinoma and normal renal tissue adjacent to carcinoma and its correlation with tumor pathological grade and clinical stage.METHODS A total of 70 patients with clear cell renal cell carcinoma from Jun.2012 to Jun.2014 in our hospital were collected and analyzed.All the patients underwent radical nephrectomy or partial nephrectomy in our hospital.The specimens were made into renal clear cell carcinoma tissue microarray and adjacent normal renal tissue microarray,with 70 cases in each group.All patients were followed up for 3 years.The immunohistochemical staining was used to compare the difference of N-WASP expression between two kinds of chips.The correlation between the expression level of N-WASP and tumor pathological grade and clinical stage was analyzed.Univariate analysis and multivariable analysis of survival rate in patients with clear cell renal cell carcinoma were carried out.RESULTS Cancer tissue N-WASP expression level was（75.54±8.48）,which was significantly lower than（121.85±10.46）（P〈0.05）.After 3 years of follow-up,the survival rates of weak and moderate level of N-WASP was 80.95%,which was higher than 25.00% strong level of N-WASP（P〈0.05）.Age and tumor diameter were the related factors of survival in patients with clear cell renal cell carcinoma（P〈0.05）.N-WASP expression level was an independent factor of survival in patients with clear cell renal cell carcinoma（P〈0.05）.CONCLUSION The expression level of N-WASP in renal clear cell carcinoma was decreased,which was an independent predictor of survival in patients with clear cell renal cell carcinoma.
