摘 要:目的用树脂去除香菇多糖粗品中的蛋白质,纯化香菇多糖。方法以香菇多糖得率和蛋白质去除率为指标,比较5种不同种类树脂(D303、D315、HZ816、HZ801、FPA98)对香菇多糖粗品吸附的情况,并通过单因子试验与正交试验优化树脂去除香菇多糖中蛋白质的工艺参数。结果强碱性阴离子树脂FPA98吸附效果最好,在p H值为9.0,上样质量浓度为2 mg/ml,上样量为每毫升树脂20 mg,流速为1 BV/h的工艺条件下,香菇多糖提取液中的蛋白质去除率可达70%以上,多糖保留率为80%以上。结论此工艺操作简单,环保,具有较好的实用性。Objective To remove the proteins from crude lentinan by resin adsorption method. Methods Based on the index of lentinan yield and protein removal rate, 5 different kinds of resins(D303, D315, HZ816, HZ801 and FPA98) were compared to the adsorption of crude lentinan, and then the technological parameters of removing the protein from Lentinan were optimized by single factor experiment and orthogonal experiment. Results The strong basic anion exchange resin FPA98 was selected as the best adsorbent. The protein removal rate could reach above 70% and the lentinan retention rate was above 80% under the optimal conditions such as pH 9.0, the sample mass concentration of 2 mg/ml, the sample loading amount with 20 mg per milliliter resin, and the flow rate of 1 BV/h. Conclusion The process is simple, environmental friendly and practical.
