摘  要:目的通过研究非小细胞肺癌组织中自分泌运动因子受体(autocrine motility factor receptor,AMFR)与肝细胞生长因子受体(hepatocyte growth factor receptor,HGF-R/C-Met)的表达,探讨二者与临床病理特征的关系及其与肿瘤细胞增殖活性的关系。方法采用SP法免疫组织化学检测肿瘤组织中AMFR、C-Met和PCNA的表达,并计算增殖细胞核抗原指数(PCNA Index,PI)。结果 AMFR表达的总阳性率为54.1%,其中,低分化组AMFR阳性表达率明显高于中-高分化组;有淋巴结转移组阳性表达率明显高于无淋巴结转移组阳性表达率。C-Met的阳性率为61.3%,其中,低分化肿瘤组织中C-Met阳性表达率明显高于中-高分化组;有淋巴结转移组阳性表达率明显高于无淋巴结转移组阳性表达率;TNM分期I-IIIA期中C-Met阳性表达率为53.7%显著低于IIIB-IV组阳性表达率。此外,AMFR与C-Met的表达率呈显著正相关,AMFR、CMet阳性表达组的PI值明显高于阴性组。结论 AMFR、C-Met两者可能在非小细胞肺癌的发展、转移中起着重要的作用。Objective To study the expression of autocrine motility factor receptor (AMFR) and hepatocyte growth factor receptor (HGF-R/C-Met) in non-small cell lung cancer (NSCLC). The relationship between the expressions and cancer clinicopathologic features as well as proliferative activity was also explored. Methods The expressions of AMFR, C-Met and proliferating cell nuclear antigen (PCNA) in tumor tissue specimens were examined using S-P immunohistochemistry. The PCNA Index (PI) was calculated. Results The expression of AMFR highly correlated with C-Met. The overall positive rates were 54.1% (AMFR) and 61.3% (C-Met) in all investigated cases, where patients with poorly differentiated tumor cells and lymph node metastasis had significantly higher positivity than those with well differentiated cells and clean lymph nodes. C-Met expression was also found to correlate with TNF classification where its positive rate was 53.7% and 82.8% in I-IIIA and IIIB-IV respectively. The PCNA Indexes were significantly higher in AMFR and/or C-Met positive tumors than those negative ones. Conclusion The expression of AMFR and C-Met had strong correlation with clinicopathological features and tumor cell proliferation which may suggest they play an important role in the progression and metastasis of the cancer.
