摘　　要：目的:通过检测PNL2单克隆抗体在不同类别黑素瘤中的表达情况,探讨PNL2在黑素瘤诊断中的意义。方法:应用免疫组织化学SP法检测47例黑素瘤(黏膜黑素瘤11例,肢端黑素瘤24例,非肢端黑素瘤12例),10例正常皮肤组织中PNL-2,S-100,HMB-45的表达情况,分析它们与患者临床生物病理特征的关系。结果:黑素瘤组PNL-2阳性率为87.23%(41/47),正常皮肤的表达率为10%(1/10),差异有统计学意义(P<0.05);PNL-2检测灵敏度和特异性分别为87.23%(41/47)和90%(9/10);其在黑素瘤中的阳性率与年龄、Clark分级有关,差异有统计学意义(P<0.05),与性别、民族、有无淋巴结转移无关(P>0.05);肢端和非肢端黑素瘤中PNL2与S100、HMB45的表达差异,无统计学意义(P>0.05);黏膜黑素瘤中PNL2表达阳性率明显大于S100、HMB45(P<0.05)。结论:PNL2对黑素瘤具有高特异性,可以作为诊断黑素瘤的一种新的生物标记物,尤其对黏膜黑素瘤的诊断有一定意义。Objective To detect the expression of PNL2 in different types of melanoma and to explore the meaning of the auxiliary diagnosis of melanoma. Methods We detected the expression of PNL2,S100 and HMB45 in 47 cases of melanoma （11 of mucosa, 24 of acromegaly and 12 of non-acra ） and 10 cases of normal skin tissue using immunohistochemistry and analyzed their relationship with the clinical pathological characteristics. Results The positive rate of PNL2 was 87.23% （41/47） in melanoma group and 10%（1/10）in normal skin. The difference was statistically significant（P〈0.05）. The sensitivity and specifcity were 87.23% （41/47） and 90%（9/10） respectively.The positive rate of PNL2 in melanoma was signifcantly related with Age and Clark grade（P〈0.05） and not signifcantly with gender, nationality and the of lymph node metastasis（P〉0.05）. Conclusion PNL2 is highly specifc for melanoma and can be used as a new biomarker for the diagnosis of melanoma, especially for the diagnosis of mucosal melanoma
