摘 要:以水黄连提取物对意大利青霉菌的抑菌圈直径为考察指标,在单因素试验基础上,采用4因素3水平的响应面优化水黄连中抑菌物质的提取条件,并将最优工艺所得提取物用于柑橘活体抑菌试验。结果表明:影响水黄连抑菌效果的主次因素为:提取时间>乙醇体积分数>液料比>温度。超声波辅助提取的最佳工艺条件为:乙醇体积分数65%,时间82 min,液料比23 mL/g,温度75℃,获得的抑菌圈直径为(52.33±0.41)mm,与理论最优值52.20 mm接近;水黄连提取物对由意大利青霉菌引发的柑橘青霉病有显著的防治作用,EC_(50)为59.53 mg/mL。Penicillium italicum was used as the tested strain. According to the single-factor experiments, a fourvariable, three-level Box-Benhnken Design( BBD) matrix was applied to optimize the extraction conditions of antifungal substance from Thalictrum smithii B. by a response surface methodology. The control efficacy of the TSB extract was tested in vivo against blue mold in citrus fruit. The statistical analysis showed that the factors influencing the extracting antifungal substance were in the order: ultrasound extraction timeethanol concentrationliquid-to-solid ratio temperature. The optimum extraction conditions were as following: ethanol concentration of65%, extraction time of 82 min, liquid-to-solid ratio of 23 mL/g and temperature of 75 ℃. Under the above extraction conditions, the average value of inhibitory zone diameter was( 52.33 ±0.41)mm, well matched with the predicted value of 52.20 mm. The TSB extract had significant efficacy in controlling blue mold in citrus fruit and the EC_(50) value was 59.53 mg/mL in vivo test.
