摘 要:目的:研究酸枣仁汤对慢性睡眠剥夺老年失眠大鼠视交叉上核c-fos及nNos表达的影响。方法:利用"多平台水环境法"建立老年大鼠慢性睡眠剥夺模型,将实验大鼠随机分为4组,环境对照组、慢性睡眠剥夺模型组、舒乐安定组以及酸枣仁汤组。用透射电镜方法观察大脑下丘脑视交叉上核部位形态学改变,免疫组织化学方法观察睡眠剥夺21 d后各组大鼠视交叉上核c-fos、nNos表达的变化,采用RT-PCR法检测大鼠视交叉上核c-fos、nNos的表达。结果:与环境对照组相比较,慢性睡眠剥夺模型组c-fos活性明显升高(P<0.05),nNos明显升高(P<0.05),脂褐素(Lipo)增多增大,扩张肿胀的高尔基体(Golgi)囊泡明显增多;与慢性睡眠剥夺模型组比较,酸枣仁汤组c-fos的表达显著下降(P<0.05),nNos明显下降(P<0.05),脂褐素显著减少减小。结论:酸枣仁汤能够调控大脑视交叉上核c-fos、nNos的表达,有效调节老年睡眠剥夺大鼠的昼夜节律。Objective:To study the influence of Suanzaoren Decoction on suprachiasmatic nucleus c-fos and nNos expressions of chronic sleep deprivation senile insomnia rats.Methods:By using the platform for water environmental law,older rats with chronic sleep deprivation model was established.The experimental rats were randomly divided into 4 groups:environmental control,chronic sleep deprivation model group,estazolam group and Suanzaoren Decoction group.Use transmission electron microscopy(sem) method to observe the hypothalamus suprachiasmatic nucleus brain regions morphology change,immunohistochemical method to observe sleep deprivation after 21 days of groups' suprachiasmatic nucleus c-fos and nNos expressions.RT-PCR method was to detect suprachiasmatic nucleus c-fos and nNos expressions.Results:Compared with the environment in the control group,chronic sleep deprivation model group's c-fos activity significantly decreased(P〈0.05),nNos increased significantly(P〈0.05) and Lipo increased.The expanded and swelling Golgi body had obviously increased Golgi vesicle.Compared with chronic sleep deprivation model group,Suanzaoren Decocion group' s c-fos expression increased significantly(P〈0.05) and nNos significantly decreased(P〈0.05) and Lipo significantly reduced.Conclusion:Suanzaoren Decoction can control the brain suprachiasmatic nucleus' c-fos and nNos expressions,effectively regulate the elderly circadian rhythm sleep deprivation in rats.
