摘 要:目的研究索骨丹中的抗氧化活性成分。方法通过DPPH自由基清除实验,首次对索骨丹乙醇浸提物的不同部位进行抗氧化活性筛选。通过多种色谱技术对抗氧化活性较强的部位进行分离纯化,结合理化性质和波谱分析鉴定其结构,再测试各化合物的抗氧化活性。结果乙酸乙酯和甲醇部位的抗氧化性较明显,从中分离到5个化合物,分别是没食子酸(1)、儿茶素(2)、胡萝卜苷(3)、正丁基-β-D-吡喃果糖苷(4)、异槲皮素苷(5)。化合物4和5首次从索骨丹中分离得到,且化合物4首次从鬼灯檠属植物中分离得到。与阳性对照Vc相比,化合物1,2和4均表现出较强的抗氧化活性。结论没食子酸、异槲皮素苷和儿茶素均有较强的抗氧化活性,阐明了索骨丹抗氧化的物质基础。Objective To study the chemical constituents with anti-oxidant activity from Rodgersia aesculifolia.Methods The DPPH free radical scavenging experiment is used for screening the anti-oxidant active fractions of ethanol extract.The compounds from the anti-oxidant active fractions are isolated and purified by variety of chromatography methods.The structures are identified by physicochemical properties and spectra analysis.Results Ethyl acetate fraction and methanol fraction showed significant anti-oxidant activity.Five compounds are obtained and identified as gallic acid(1),catechin(2),daucosterol(3),n-butyl-β-D-fructopyranoside(4),isoquercitrin(5).Compounds 4 and 5 are isolated from the plant of Rodgersia aesculifolia for the first time and compound 4 is first obtained from the genus.Compared with positive control Vc,compounds 1,2 and 4 showed strong anti-oxidant activity.Conclusion The anti-oxidant activities of gallic acid,isoquercetin and catechin are strong,which elucidate the material basis of anti-oxidant protein.
