摘  要:目的观察锌指转录因子2(zinc-finger transcription factor-2,SNAI2)在人脑胶质瘤中的表达及其对胶质瘤细胞增殖转移能力的影响。方法收集37例恶性胶质瘤患者的临床资料及肿瘤组织标本。采用反转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)技术检测组织中SNAI2的表达水平,分析其表达与患者临床病理资料间的关系。在体外实验中,构建稳定干扰SNAI2(small interfering RNA SNAI2,shSNAI2)的胶质瘤细胞株,通过噻唑蓝比色法检测细胞增殖能力,划痕实验检测细胞侵袭转移能力,Western blot检测E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)及紧密连接蛋白(Zo-1)的表达,检测细胞上皮间充质转化(epithelial mesenchymal transition,EMT)的水平。结果高级别脑胶质瘤中SNAI2的表达显著高于低级别脑胶质瘤、良性脑膜瘤及正常脑组织,且低级别脑胶质瘤SNAI2的表达显著高于良性脑膜瘤及正常脑组织,差异均有统计学意义(P<0.05)。Spearman相关分析结果显示SNAI2的表达与患者性别、年龄及体质量不相关,与患者肿瘤大小及胶质瘤细胞分级呈正相关(P<0.05)。MTS实验结果示各组细胞随时间增加增殖能力增强,且shSNAI2细胞系相比对照细胞系增殖能力显著减弱(P<0.05)。划痕修复实验结果示相比对照细胞系转移能力明显减弱(P<0.05)。Western blot结果显示shSNAI2细胞系E-cadherin及Zo-1表达增高,Vimentin表达降低,差异均有统计学意义(P<0.05)。结论 SNAI2在人脑胶质瘤中高表达,且与患者肿瘤的大小和分级密切相关,其能够促进胶质瘤细胞的增殖、转移及EMT过程的发生,是潜在的促癌基因。Objective To investigate the mRNA expression level of zinc-finger transcription factor-2(SNAI2)gene in clinical tissues,and to identify the effect of SNAI2 on the proliferation and invasion of glioma cancer cells U251.Methods A total of 37 cases of patients with malignant glioma was selected in our hospital.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the mRNA level of SNAI2 in tissues and to analyze the relationship between the expression and the clinical pathological data.In vitro,we set up a stable SNAI2 interference(shSNAI2)of glioma cells.The ability of cell proliferation was detected by MTT colorimetric assay(MTS).The ability of cell invasion and metastasis was detected by scratch assay,Western blot was used to detect the expression of E-cadherin,vimentin and Zo-1,related with epithelial mesenchymal transition(EMT).Results The expression of SNAI2 in high-grade gliomas was significantly higher than that in low grade gliomas,benign meningiomas and normal brain tissue,and the expression of SNAI2 in low grade gliomas was significantly higher than that in benign meningiomas and normal brain tissue,and the difference was statistically significant(P<0.05).Spearman correlation analysis was used to compare the relationship between the expression of SNAI2 and the clinical pathological data of patients,and the results show that the expression of SNAI2 and the patient's gender,age and body weight were not statistically related.There was a positive correlation between tumor size and tumor size and tumor grade(P<0.05).The results of MTS experiment showed that the proliferation ability of cells in each group increased with time,and the proliferation ability of shSNAI2 cell line was significantly lower than that of control cell line(P<0.05).The results of scratch repair showed that the ability of cell migration of shSNAI2 cell line was significantly less than that of control cell line.The results of Western blot showed that the expression of E-cadherin and Zo-1 in shSNAI2 cell line was increase
