摘  要:目的研究Ras相关区域家族5(NORE1)基因过表达对胃癌细胞迁移、侵袭的影响及其作用机制。方法采用前瞻性研究将胃癌细胞分为NORE1过表达组和对照组,分别转染人的胃癌AGS细胞和SGC7901细胞,Westren blot检测细胞转染效率;Transwell实验检测细胞迁移和侵袭的能力;Westren blot检测胃癌AGS细胞和SGC7901细胞中星形胶质细胞上调基因1(AEG-1)和基质金属蛋白酶9(MMP-9)的蛋白表达水平。结果转染NORE1过表达质粒后,胃癌AGS细胞和SGC7901细胞中NORE1表达量显著高于对照组(P<0.05)。胃癌细胞AGS、SGC7901的迁移、侵袭能力减弱,与对照组具有显著差异(P<0.05)。与对照组相比,NORE1过表达组胃癌细胞AGS、SGC7901中AEG-1蛋白和MMP-9蛋白的表达量显著下降(P<0.05)。结论胃癌细胞中NORE1基因可以通过调节AEG-1蛋白和MMP-9蛋白的表达量影响其迁移和侵袭能力。Objective To investigate the effects of novel Ras effector 1( NORE1) overexpression on the migration,invasion of AGS cell and SGC7901 cell,and to explore the related molecular mechanisms. Methods NORE1 overexpression plasmid and negative control were transiently transfected into AGS cell and SGC7901 cell using Lipofectamine TM2000. Western blotting was performed to measure the expression of AEG-1 and MMP-9. The cellular capabilities of migration,invasion were detected by Transwell assay and vitro angiogenesis experiment respectively. Results The expression of NORE1 in AGS cells and SGC7901 cells were significantly higher than that in control group( P 〈 0. 05). The migration and invasion of AGS and SGC7901 were significantly lower than those of the control group( P 〈 0. 05). Compared with the control group,the expression of AEG-1 protein and MMP-9 protein in AGS and SGC7901 of NORE1 overexpression group were significantly decreased( P 〈 0. 05). Conclusion NORE1 in both AGS cell and SGC7901 cell can affect cell migration and invasion by regulating the expression of AEG-1 and MMP-9.
