摘 要:毕赤酵母是真核细胞常用表达系统之一,具有细胞密度高、纯化方法简单、转录后修饰功能完善等优点。作为重要的调控元件,表达系统中启动元件的功能强弱与表达效率的高低密切相关。目前,毕赤酵母表达系统中常用的启动元件分为三类,分别是以p AOX为代表的甲醇诱导型、以p GAP为代表的非甲醇诱导型,还有一些新型工业酵母启动子也是理想的表达系统启动元件。充分了解这些酵母启动子的最新研究进展,可以为工业酵母表达系统的优化提供重要的发展思路。Pichia pastoris is one of the most common expression systems for eukaryotic cells, with the advantages of high cell density, simple purification methods, and the perfect function of post-transcriptional modification. As an important regulatory el-ement, the function of the promoter element in the expression system is closely related to the level of expression efficiency. At present, the promoter elements commonly used in Pichia pastoris expression system are divided into three types: the methanol inducible promoter represented by pAOX , non-methanol inducible promoter like pGAP, and some new industrial yeast promot-ers are also ideal promoter elements. To fully understand the latest research progress of these yeast promoters could provide an important development idea for the optimization of industrial yeast expression system.
