星形细胞上调基因-1与结肠癌临床病理及预后的相关性研究

摘　　要：目的探讨星形细胞上调基因-1(astrocyte elevated gene-1,AEG-1)与结肠癌临床病理及预后的相关性。方法选取南方医科大学新塘医院外一科2008年1月至2010年12月收治的62例结肠腺癌患者病例资料。采用免疫组织化学的方法检测癌旁正常组织和结肠癌组织中AEG-1蛋白的表达水平,探讨结肠癌组织中AEG-1与临床病理特点和预后的关系。结果结肠肿瘤组织中AEG-1免疫组化阳性表达率显著高于癌旁正常结肠组织[67.7%vs.30.0%,P=0.03]。结肠癌组织AEG-1表达与肿瘤大小、T分期和TNM分期显著相关(均P<0.05)。AEG-1表达阳性的结肠癌患者总体预后较AEG-1阴性者差。AEG-1表达阳性患者和表达阴性患者的5年总体生存率分别为30.5%和74.3%(P=0.004)。多因素分析结果显示AEG-1阳性表达是结肠癌患者预后不佳的独立因素。结论 AEG-1阳性表达与结肠癌患者的临床病理及较差的生存预后密切相关。Objective To investigate the correlation between the astrocyte elevated gene-1 and clinical pathology and prognosis of colon cancer. Methods 62 patients with colon adenocarcinoma from January2008 to December 2010 were selected from the xintang hospital of southern medical university. By immunohistochemical analysis, we investigated AEG-1 expression in adjacent normal tissue and colon carcinoma. The relationship between AEG-1 and clinicopathological features and prognosis of patients with colon carcinoma were studied. Results We found that positive AEG-1 expression were more common in colon carcinoma compared with adjacent normal tissue[67.7% vs. 30.0%,P=0.03 ]. We revealed that AEG-1 expression was markedly correlated with the tumor size, T classification and TNM stage in the colon cancer patients(all P <0.05). Furthermore,those patients with high AEG-1 levels had shorter survival time than the ones with low AEG-1 levels(5-year survival rate: 30.5% vs. 74.3%, P=0.004). Multivariate Cox regression analysis revealed that the overexpression of AEG-1 protein was the independent prognosis factor for worse overall survival in our study. Conclusion High AEG-1 expression correlates with worse clinicopathological features and poor overall survival in the colon carcinoma patients.
