摘 要:目的探讨micro RNA-182(mi RNA-182)在人肺腺癌组织中的表达,以及与患者临床特征及预后的相关性。方法收集术后或穿刺活检病理证实的人肺腺癌新鲜组织98例,以及其癌旁正常肺组织和良性病变旁正常肺组织98例,实时荧光定量聚合酶链反应检测mi RNA-182的相对表达量,分析其与临床特征及生存预后的关系,用ROC曲线评估mi RNA-182对人肺腺癌的诊断能力。结果 mi RNA-182在人肺腺癌组织相对表达量为(0.8001±0.2112),在正常肺组织相对表达量为(0.2721±0.1216),人肺腺癌组织中的表达高于正常肺组织(P<0.05);mi RNA-182的表达水平与TNM分期、淋巴结转移、远处转移及肿瘤分化相关(P<0.05)。ROC曲线分析发现,当界值设置为0.52时,mi RNA-182在人肺腺癌中的表达特异性为95.918%,敏感性为91.837%,阳性预测值为95.745%。结论 mi RNA-182在人肺腺癌组织中表达量高于正常肺组织,有可能成为人肺腺癌诊断、预后评估的重要指标。Objective To study the level of micro RNA-182(mi RNA-182) in human lung adenocarcinomas,and to evaluate its correlations with clinical features and prognosis of the patients with lung adenocarcinomas.Methods The fresh human lung adenocarcinoma tissues and their paired adjacent normal lung tissues of 98 cancer patients, and normal lung tissues of 98 cases with benign lesions were collected. All tissues of surgery or biopsy were confirmed by pathology. mi RNA-182 was measured by q RT-PCR. The correlations of mi RNA-182 with clinical features and prognosis of human lung adenocarcinomas were analyzed. The ROC curve was used to evaluate the ability of mi RNA-182 in diagnosis of human lung adenocarcinomas. Results The level of mi RNA-182 in the human lung adenocarcinomas(0.8001 ± 0.2112) was significantly higher than that in the normal lung tissues [(0.2721 ± 0.1216), P < 0.05]. The level of mi RNA-182 in human lung adenocarcinomas was correlated with TNM stage, lymphatic metastasis, distant metastasis and tumor differentiation(P < 0.05).After further analysis by ROC curve, the results showed that when the cut-off value was set to 0.52, the specificity, the sensitivity and the positive predictive value of mi RNA-182 in human lung adenocarcinomas were 95.918%, 91.837% and 95.745% respectively. Conclusions Level of mi RNA-182 in human lung adenocarcinomas is significantly higher than that in normal lung tissues, it may be an important index for the diagnosis of human lung adenocarcinomas and prognosis evaluation.
