摘  要:目的:探讨高脂饮食对大鼠肾组织中炎症因子抑瘤素-M(OSM)表达的影响。方法:将健康雄性Wistar大鼠随机分为普通饮食组、高脂饮食4周组、高脂饮食8周组、高脂饮食12周组4组,应用蛋白免疫印迹技术检测各组大鼠肾组织中OSM的表达变化,ELISA法检测各组大鼠血清中OSM表达,生化方法检测肾功能、血脂及24 h尿蛋白定量,对各指标与OSM表达进行相关性分析。结果:高脂血症大鼠血清中OSM表达较对照组明显增高(P<0.05),且随着高脂饮食时间的延长,OSM表达亦明显升高。生化检测提示,随高脂饮食时间的增加,肾功能、血脂、24 h尿蛋白定量逐渐增加,OSM表达与肾功能、血脂、24 h尿蛋白定量呈显著正相关(P<0.05)。结论:高脂环境下大鼠肾组织OSM表达明显增加,且与肾功能、血脂、24 h尿蛋白定量明显相关,提示OSM表达很可能参与高脂致肾损害的病理过程。Objective: To investigate the effect of high lipid diet on the expression of inflammatory factor oncostatin- M (OSM) in rat kidney. Methods: The healthy male Wistar rats were randomly divided into 4 groups : normal diet group, high fat diet group fed for 4 weeks, high fat diet fed for 8 weeks and high fat diet for 12 weeks. The expression of OSM in renal tissue was detected by immunoblotting. The expression of OSM in serum was detected by ELISA. The biochemical method was used to detect renal function, blood lipid and 24-hour urine protein, and the correlation between OSM expression and each index was analyzed. Results: The expression of OSM in hyperlipidemia rats was significantly higher than that in control group ( P 〈 0. 05 ), and with the prolongation of high-fat diet, the expression of OSM also increased significantly( P 〈 0.05 ). There was a significant and positive correlation between OSM and renal function, blood lipid and 24- hour urine protein ( P 〈 0.05 ). Conclusion: The expression of OSM in high lipid rats increases significantly, which is correlated with renal function, blood lipid and 24-hour urine protein, suggesting that OSM expression is likely to be involved in the pathogenesis of renal injury induced by high lipid.
