摘　　要：目的探讨4'-甲醚-黄岑素(4'-MS)对人绒毛膜癌细胞凋亡的影响。方法采用流式细胞术、实时定量PCR法及RT-PCR技术体外观察4'-MS对JAR细胞的影响。结果流式细胞术分析,加药组细胞凋亡率随4'-MS浓度的增加而逐渐升高,G2/M期细胞比例显著升高。20、40 mg/ml 4'-MS作用48 h后c-myc及h TERT mRNA的表达量明显低于对照组(P<0.01)。RT-PCR结果显示,20 mg/ml和40 mg/ml 4'-MS作用后,人绒毛膜癌JAR细胞中Survivin mRNA表达显著下降,而Caspase 9、Caspase 3及p38 MAPK mRNA表达显著上升(P<0.05)。结论绒毛膜癌JAR细胞在4-MS的作用下发生凋亡的机制与降低c-myc及h TERT mRNA的表达、上调Caspase 9表达、Caspase 3表达、激活p38 MAPK信号通路及抑制Survivin表达相关。Objective To explore the effect of 4＇-methyl ether-scutellarein（4＇-MS） on apoptosis of human choriocarcinoma cells（ JAR cells）. Methods Flow cytometry,real-time quantitative PCR,and RT-PCR were used to observe the effect of 4＇-MS on JAR cells in vitro. Results The results of flow cytometry showed that the apoptotic rate of JAR cells in drug group gradually increased with the increase of 4＇-MS concentration,the cells at G2/M phase increased significantly. The expression levels of c-myc and h TERT mRNA at 48 hours after treatment of 4＇-MS（ 20 mg/ml and 40 mg/ml） were statistically significantly lower than those in control group（ P0. 01）.The results of RT-PCR showed that the expression level of Survivin mRNA in JAR cells after treatment of 4＇-MS（ 20 mg/ml and 40 mg/ml）decreased,while the expression levels of Caspase 9,Caspase 3,p38 MAPK mRNA increased significantly（ P0. 05）. Conclusion The apoptosis of JAR cells after treatment of 4＇-MS is correlated with decrease of c-myc and h TERT mRNA expression levels,up-regulation of Caspase 9 and Caspase 3,activation of p38 MAPK signal pathway,and inhibition of Survivin expression.
