三氧化二砷对人子宫内膜癌细胞裸鼠模型中Caspase-3表达的影响

摘　　要：目的通过建立人子宫内膜癌细胞HEC-1-B裸鼠模型,检测不同浓度三氧化二砷(As_2O_3)对移植瘤细胞Caspase-3表达的影响,并探讨As_2O_3的作用机制。方法建立人子宫内膜癌HEC-1-B细胞皮下移植瘤模型,按编号随机分为实验组即As_2O_3低(1.0 mg/kg)、中(2.5 mg/kg)、高(5.0 mg/kg)剂量组、空白对照生理盐水(NS)组及阳性对照顺铂组(5.0 mg/kg),连续腹腔给药8天后,通过流式细胞仪及实时荧光定量RT-PCR检测移植瘤内Caspase-3相对表达量。结果与生理盐水组比较,低、中浓度As_2O_3及顺铂均可上调Caspase-3的表达(P<0.05),但高浓度组上调不明显(P>0.05)。与顺铂组比较,As_2O_3低剂量组作用最明显,差异有统计学意义(P<0.05);但与中、高浓度组比较,差异无统计学意义。结论低浓度As_2O_3能上调人子宫内膜癌细胞内Capase-3的表达,且优于顺铂,可能是三氧化二砷诱导细胞凋亡的作用机制之一。Objective To study the effect of arsenic trioxide （ As2 O3 ） on expression of Caspase-3 in nude mice models of human endometrial carcinoma cell lines （Hec-1B）.Methods The nude mice models of endometrial carcinoma cell lines （Hec-1B）were randomized into 5 groups via number,then they were treated with NS,DDP,increasing concentrations of As2O3（1.0 mg/kg,2.5 mg/kg,5.0 mg/kg） by intraperitoneal injection.The Caspase-3 expression was detected by flow cytometry and real-time PCR.Results Compared with NS group,the As2O3（1 mg/kg、2.5 mg/kg）groups and DDP group increased significantly the ex-pression of Caspase-3（P〈0.05）.But As2O3（5 mg/kg）group hadn＆#39;t distinguished effection.Compared with DDP group,the As2O3（1 mg/kg）group was statistically significant（P〈0.05）,but the As2O3（2.5 mg/kg、5 mg/kg）groups weren＆#39;t.Conclusion （1 mg/kg）As2O3 can increase the expression of Caspase-3,and was superior to DDP,the action may be one of mechanisms of ar-senic trioxide inducing apoptosis.
