摘  要:目的观察肿瘤坏死因子α(TNF-α)在SOD1-G93A转基因小鼠不同时期腰髓、胸髓中的变化,探讨TNF-α与肌萎缩侧索硬化症(ALS)小鼠模型疾病进展的关系。方法采用酶联免疫吸附实验(ELISA)、RT-PCR方法,观察不同疾病时期SOD1小鼠胸髓、腰髓中TNF-α水平及mRNA变化。结果 SOD1小鼠在症状早期即有TNF-α分泌增加,随病情进展TNF-α水平增加。结论随病情进展TNF-α分泌水平增加,TNF-α介导的神经系统炎症在ALS发病机制中起比较关键的作用。Objective To observe the change of TNF-αin the lumbar and thoracic spinal cord of SOD1-G93 Atransgenic mice at different stages and discuss the relationship between and progression of amyotrophic lateral sclerosis(ALS).Methods The level of TNF-αand TNF-αmRNA was analyzed by ELISA and RT-PCR at presymptomatic,symptomatic and end stages of SODl-G93 Atransgenic mice.Results Increasing TNF-αand TNF-αmRNA appeared at presymptomatic stage in the lumbar and thoracic spinal cord of SODl-G93 Atransgenic mice,and became obvious at symptomatic stage and reached the maximum at end stage.Conclusion TNF-αproliferates markedly with the progression symptom of SODl-G93 Atransgenic mice.TNF-αmediated neuroinfalammation,play an important role in the pathophysiology of ALS.
