摘  要:目的:探讨Shugoshin1(Sgo1)对于非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞株H1299增殖的影响。方法:用Western blot及RT-PCR检测Sgo1在NSCLC细胞株H1299中的表达。成功构建Sgo1siRNA质粒,经慢病毒包装转染H1299细胞,利用Western blot及RT-PCR鉴定转染效力。用单层细胞生长实验和平板克隆形成实验观察Sgo1对H1299细胞株增殖及克隆形成能力的影响。结果:在NSCLC细胞株H1299中,Sgo1表达明显高于人成纤维细胞株。下调Sgo1的表达后,H1299细胞株的增殖能力和克隆形成能力均明显减低。结论:Sgo1的表达与NSCLC细胞株H1299的增殖密切相关,有可能成为肺癌治疗的一个新靶点。Objective: To explore the role of Shugoshinl ( Sgol ) in human non - small cell lung cancer (NSCLC) H1299. Methods :By RT- PCR and Western blot,the expression of Sgol in NSCLC cell lines was measured. We suc- cessfully constructed a lentivirus vector mediating RNAi targeting of Sgol ( LV - Sgol siRNA) and proved that it could suppress the expression of Sgol gene in H1299 cells. The monolayer growth assay and colony formation assay were used to determine the growth of NSCLC cell lines. Results: RT - PCR and Western blot showed that Sgol was highly expressed in H1299. RNA interference of Sgol by the LV - Sgol siRNA construct significantly decreased Sgol protein expression and inhibited the growth and ability of forming colonies of H1299 cells, but lentivirus vector control had no effect on H1299 cells. Conclusion:Sgol could inhibit the growth of lung cancer cells. It might provide a novel therapeutic target for the treatment of lung cancer.
