摘 要:目的:探讨RNA干扰MAGE-A3基因对白血病细胞增殖的影响。方法:构建MAGE-A3干扰基因重组质粒载体,转染K562白血病细胞系,观察各组白血病细胞MAGE-A3基因表达及白血病细胞增殖情况。结果:对照组及空白质粒组白血病细胞MAGE-A3基因表达明显增高,而重组质粒组MAGE-A3基因表达显著降低;重组质粒组白血病细胞在24 h、48 h及72 h的增殖活性逐渐减弱,且重组质粒组白血病细胞的增殖活性明显低于对照组及空白质粒组白血病细胞。结论:RNA干扰能显著抑制MAGE-A3基因表达并进一步抑制白血病细胞的增殖活性。Objective: To investigate the influence of MAGE - A3 gene on proliferation of leukemia cells by RNA interference. Methods : Detecting the expression of MAGE - A3 gene and proliferation of K562 leukemia cells which transfected by recombinant plasmid constructed with MAGE - A3 interference gene. Results : The expression of MAGE -A3 gene was significantly elevated in control and blank plasmid groups ,while its expression was remarkable low in recombinant plasmid group. The proliferation activity of leukemia cells in recombinant plasmid group gradually dimin- ished in 24 h,48 h and 72 h after transfection, and the proliferation activity of leukemia cells in recombinant plasmid group was significantly low in comparison with control and blank plasmid groups. Conclusion: The expression of MAGE -A3 gene was remarkable inhibited by RNA interference ,followed by suppressed proliferation activity of leu- kemia cells.
