疏肝化浊方对H22荷瘤小鼠的抗肿瘤作用及机制

摘　　要：目的:观察疏肝化浊方对H22荷瘤小鼠抗肿瘤的效果,研究可能存在的作用机制。方法:将50只荷瘤小鼠分为模型组、疏肝化浊方低、中、高剂量组及阳性对照组(5-FU),比较各组小鼠抑瘤率、脾脏指数、胸腺指数、淋巴T/B细胞增殖活性、IL-2及TNF-α含量变化情况;RT-PCR测定VEGF、TGF-β1及NF-κB基因表达情况,HE染色观察各组小鼠肝脏组织病理学变化情况。结果:疏肝化浊方可有效提高抑瘤率、增加脾脏及胸腺指数,增强免疫细胞的增殖活性,提高IL-2含量及TGF-β1的表达,降低TNF-α含量及VEGF、NF-κB基因的表达。结论:疏肝化浊方对H22荷瘤小鼠具有较好的抗肿瘤效果,其作用机制可能与提高小鼠免疫功能、降低肿瘤内部血管形成、抑制肿瘤细胞的分化有关。Objective： To observe the effect of Shugan Huazhuo fang on anti - tumor effect of H22 tumor - bearing mice, and to study the possible mechanism of action. Methods： Fifty tumor - bearing mice were divided into model group, low - dose, medium, high - dose group and positive control group （5 - FU ）. The changes of tumor growth rate, spleen index, thymus index,lymphocyte T/B cell proliferation activity, IL- 2 and TNF-or levels were compared. The expression of VEGF,TGF - β1 and NF - κB were detected by RT - PCR. HE staining was used to observe the his- topathological changes of liver in mice. Results： Shugan Huazhuo fang can effectively increase the rate of tumor sup- pression, spleen, thymus index, and proliferation activity of immune cells, increase the content of IL -2 and the ex- pression of TGF - β1, reduce the content of TNF - α and the expression of VEGF, NF - κB genes. Conclusion：Shu- gan Huazhuo fang has a good anti - tumor effect on H22 tumor - bearing mice, and its mechanism may be related to improving the immune function of mice, reducing tumor internal angiogenesis and inhibiting the differentiation of tumor cells.
