摘 要:目的探讨miR-106a、miR-376c、miR-377、miR-92a和miR-130在5种白血病细胞系中的表达水平及其临床意义。方法分别提取5种白血病细胞系small RNA,利用miRNA特异性的茎环引物进行反转录,合成c DNA,利用Real-Time PCR检测各白血病细胞系miRNA分子的表达情况。结果与对照组比较,5种miRNAs在NAML^(-1)细胞系中表达均上调(P<0.05);在KOCL-33细胞系、KOCL-44细胞系、KOPB-26细胞系和KOCL-50细胞系中,miR-106a表达均上调,miR-92a表达均下调(P均<0.05);miR-376c在KOCL-33、KOCL-44细胞系中表达上调,在其余2种细胞系中表达下调(P<0.05);miR-377在3种细胞系中表达均上调(P<0.05),在KOCL-33细胞系中表达差异无统计学意义(P>0.05);miR-130在KOCL-33、KOCL-44和KOCL-50细胞系中表达均下调(P均<0.05)。结论 miR-106a、miR-376c、miR-377、miR-92a和miR-130在KOCL-33、KOCL-44、KOPB-26、KOCL-50及NAML^(-1) 5种白血病细胞系中表达量均有变化,表明这些miRNAs在此类白血病发生、发展过程中可能发挥一定作用。Objective To investigate the expression and its clinical significance of miR-106 a,miR-376 c,miR-92 a and miR-130 in five leukemia cell lines with KOCL-33,KOCL-44 etc,and to provide a theoretical basis for the further research in the mechanism of leukemia. Methods Small RNA was extracted in different leukemia cell lines. After using the stem-loop reverse transcription primers to reverse transcribed into c DNA,the expression of miR-106 a,miR-376 c,miR-377,miR-92 a and miR-130 was detected by Real-Time PCR.Results Compared with control group,the expression of 5 miRNAs in the NAML^-1 cell line was significantly up-regulated(P〈0.05). In leukemia cell line of KOCL-33,KOCL-44,KOPB-26 and KOCL-50 group,the expression of miR-106 was up-regulated but miR-92 a was down-regulated. The expression of miR-376 c was up-regulated in KOCL-33 and KOCL-44 cell lines,but it was down-regulated in the other two cell lines.Mi R-377 was high expression in three cell lines,and no significant difference was found in KOCL-33 cell line(P〈0.05). The expression of miR-130 was increased significantly in KOCL-33,KOCL-44 and KOCL-50 cells except it was decresed significantly in KOCL-44 cell line(P〈0.05). Conclusion The expression of miR-106 a,miR-376 c,miR-377,miR-92 a and miR-130 changed significantly in the five different cell lines with leukemia.It indicated that the miRNAs may play an role in the developing process of the leukemia.
