摘  要:目的筛查早期、进展期胃癌和健康人群血清中差异表达的蛋白,以期发现与胃癌有关的蛋白标志物。方法用同位素标记相对和绝对定量技术(iTRAQ)联合串联液相色谱/质谱联用(LC-MS/MS)检测早期与进展期胃癌患者血清,筛选关键性差异表达腺苷酸核糖基化因子(ARF-1)蛋白,利用免疫组化进行胃癌组织蛋白差异验证。结果共发现119个差异蛋白质(53个上调,66个下调)。免疫组化结果显示进展期胃癌组织中ARF-1蛋白表达量高于早期胃癌组与癌旁正常组,早期胃癌组ARF-1蛋白表达量高于癌旁正常组(P<0.05)。结论初步筛选了胃癌血清中蛋白表达谱,经免疫组化验证ARF-1表达水平与血清一致。Objective To identify protein markers related to gastric carcinoma,We screened the differentially expressed proteins in serum of the early and advanced gastric carcinoma as well as healthy people.Methods Using isotopic labeling by the relative and absolute quantification(iTRAQ) combined liquid chromatography tandem mass spectrometry(LC-MS/MS) proteomics technology the differentially expressed protein ARF-1 was screened. These proteins between gastric cancer tissue and normal tissue adjacent to carcinoma were validated using. Results We got a total of 119 differentially expressed proteins,with 53 proteins up-regulated and 66 proteins down-regulated. Immunohistochemistry showed that ARF-1 expression in advanced gastric carcinoma was significantly higher than that in normal tissues and the early group. Besides,the ARF-1 in the early gastric cancer group was significantly higher than that in the non-tumor group(P〈0.05).Conclusion The serum expression of protein in gastric cancer was preliminarily screened,and,the serum expression level of ARF-1 was consistent with that by immunohistochemistry.
