达沙替尼抑制乳腺癌增殖及侵袭转移能力的初步研究

摘　　要：目的:探索达沙替尼对三阴性乳腺癌细胞增殖、侵袭迁移能力的影响及相关机制。方法:选用人三阴性乳腺癌Hs578T细胞系,采用PrestoBlueTM细胞活性增殖实验及琼脂糖克隆形成实验检测达沙替尼对乳腺癌细胞增殖及克隆形成能力的影响;Transwell侵袭实验及划痕实验研究达沙替尼对乳腺癌细胞侵袭及迁移能力的影响,并对其作用机制进行初步研究。结果:细胞增殖及克隆形成实验结果显示达沙替尼可明显抑制三阴性乳腺癌细胞的增殖及克隆形成能力。Transwell侵袭实验及划痕实验结果显示达沙替尼可使乳腺癌细胞的侵袭及迁移能力明显降低。Western Blot结果显示,达沙替尼可明显抑制乳腺癌细胞p-STAT3表达水平。结论:达沙替尼可能通过抑制STAT3信号传导通路抑制三阴性乳腺癌细胞的增殖、克隆形成及侵袭、迁移能力。Objective： Our study attempts to investigate the effects and the mechanism of dasatinib on the proliferation, invasion and metastasis of triple-negative breast cancer （TNBC） cel）s. Method： TNBC Hs578T cells were tested for the sensitivity to dasatinib in vitro using PrestoBlueTM reagent and mammosphere assay. Transwell assay and wound-healing assay were used to deter mine invasion and metastasis ability of Hs578T cells. The p-STAT3 and STAT3 expressions of dasa- tinibr-ltreated Hs578T cells were measured by Western blot. Results： Dasatinib exhibited growth inhibition of Hs578T cells. Mammosphere assay showed that dasatinb treatment led to less mam-mosphere formation of TNBC cells. More importantly, migration and invasion ability of Hs578T cells was also decreased by dasatinib treatment. Western blot analysis revealed that the P-STAT3 level was decreased significantly in Hs578T cells after dasatinib treatment, compared with control group. Conclusions： Dasatinib prevents the proliferation, invasion and metastasis ability of TNBC cells via inhibiting STAT3 pathway.
