摘 要:目的探讨氢质子磁共振波谱成像(~1H-MRS)测定Fmr1基因敲除小鼠(KO小鼠)脑内代谢物的变化及诊断价值。方法 23只4月龄KO小鼠(实验组)和16只野生型FVB小鼠(WT小鼠,对照组)采用3.0T高场强MRI行~1H-MRS,测量小鼠大脑皮质与海马区N-乙酰天门冬氨酸(NAA)、胆碱(Cho)和肌酐(Cr)的峰下面积,计算NAA/Cr和Cho/Cr比值并进行两样本均数t检验。结果与对照组比较,实验组小鼠NAA/Cr比值明显降低(t=-5.912,P<0.01),Cho/Cr比值明显升高(t=3.888,P<0.01)。结论~1H-MRS能定量测定KO小鼠脑内代谢物(细胞膜代谢)异常,可能为诊断及评价脆性X综合征疗效提供重要依据。Objective To investigate the changes of metabolites in brain of Fmrl knockout mice (KO mice) by 1H - MR spectroscopy ( 1H - MRS) and its application in diagnosis of Fragile X Syndrome ( FXS ). Methods 1H - MRS was performed on 23 KO mice (experimental group) and 16 wild type mice (control group) at the age of 4 months. The sub - peak areas of N - aeetyl aspartate (NAA) , choline (Cho) and creatine (Cr) in the cerebral codex and hippo- campus were measured, and the NAA/Cr and Cho/Cr ratios were calculated and compared by two - sample t - test. Results Compared with control group, the NAA/Cr ratio was significantly reduced ( t = - 5. 912, P 〈 0.0l ) , and the Cho/ Cr ratio was significantly increased (t = 3. 888, P 〈 0. 01 ) in KO mice. Conclusion 1H - MRS examination can quantitatively assess abnormal metabolites in the brains of KO mice, which may provide important clues for the diagnosis of FXS and evaluation of therapeutic efficacy.
