金水鲜联合放疗对肺腺癌A549细胞碱性成纤维细胞生长因子和低氧诱导因子-1α表达的影响

摘  要:目的观察金水鲜胶囊联合放疗对肺腺癌A549细胞碱性成纤维细胞生长因子(bFGF)和低氧诱导因子-1α(HIF-1α)蛋白及mRNA表达的影响。方法体外培养A549细胞,应用简单随机化分为4组:空白对照组、金水鲜组、放疗组以及金水鲜联合放疗组。采用噻唑蓝(MTT)法检测细胞生长抑制率,原位缺口末端标记法(TUNEL)法检测细胞凋亡,酶联免疫吸附试验(ELISA)法检测bFGF、HIF-1α蛋白的表达,实时定量聚合酶链反应(Real-time PCR)检测各组bFGF及HIF-1α mRNA的表达。结果处理组细胞的增殖在不同时间点呈现出不同程度的抑制,48 h时金水鲜联合放疗组抑制率最高(56.3%),金水鲜组最低(15.8%)。联合组抑制细胞增殖程度高于金水仙组、放疗组,差异有统计学意义(P=0.023)。凋亡实验结果表明,48 h时联合组凋亡细胞数最多,与其他组比较,差异有统计学意义(P=0.038)。各处理组A549细胞bFGF和HIF-1α蛋白的表达有所下降,其中金水鲜联合放疗组在48 h时bFGF[(157.408 0±2.830 3) pg/ml、HIF-1α(1.455 7±0.000 7) pg/ml]表达量最低,与其他各组比较,差异有统计学意义(P=0.045、0.021)。与对照组比较,各处理组bFGF、HIF-1α mRNA的表达均表现出下降趋势,在48 h时金水鲜联合放疗组表达最低(0.285 4±0.057 9、0.277 1±0.022 1),与其他各组比较,差异有统计学意义(P=0.005、0.011)。结论金水鲜联合放疗可显著抑制肿瘤细胞增殖及bFGF和HIF-1α的表达,诱导肺腺癌A549细胞凋亡。金水鲜可能通过抑制bFGF和HIF-1α的表达进而增强放疗对肺癌A549细胞的抑制作用。Objective To observe the effects of JinShuiXian capsule combined with radiotherapy on the expression of basic fibroblast growth factor (bFGF) and hypoxia inducible factor-1α (HIF-1α) in lung adenocarcinoma A549 cells. Methods A549 cells were cultured in vitro. Cells were divided into 4 groups using simple random method: control group, JinShuiXian group, radiation group and JinShuiXian combined with radiation group. The cell growth inhibition rate of each group at different time sets was monitored by methyl thiazol tetrazolium (MTT), the number of apoptotic cells in different groups was observed by TdT - mediated dUTP nick end labeling (TUNEL) , and the expression levels of mRNA and protein of HIF - 1α and bFGF were detected by real - time quantitative polymerase chain reaction ( Real - time PCR) and enzyme linked immunosorbent assay (ELISA). Results The proliferation of A549 cells in different groups at different time points showed different degrees of inhibition. Especially at 48 h, the inhibitory rate of combination group was 56. 3% , and the JinShuiXian group had the lowest rate (15.8% ). As compared with control group, JinShuiXian group and radiation group, the inhibitory rate of cell proliferation in combined group at 48 h was significantly increased (P = 0.023). Apoptosis experiments showed that the mean number of apoptotic ceils in combined group at 48 h was 32. 8, which was significantly increased as compared with other groups ( P = 0. 038 ). The expression levels of bFGF [ ( 157. 408 0±2. 830 3 ) pg/ml, P =0. 045] and HIF -1α [ ( 1. 455 7 ±0. 000 7) pg/ml, P = 0. 021 ] proteins showed a decline trend over time in each group, and those in the combined group were reduced most significantly as compared with other groups. As compared with the control group, bFGF (0. 285 4 ± 0. 057 9, P = 0. 005 ) and HIF -1α mRNA (0. 277 1 ±0. 022 1, P =0. 011 ) expression levels in the treated groups showed a downward trend, and at 48 h those in the combined gro
