摘  要:目的 探讨药物注射及手术结扎两种方法制造兔心肌梗死(MI)模型,在颈上神经节(SCG)的神经元通道特性方面的研究对比.方法 药物注射制造MI模型是利用异丙肾上腺素(ISO)腹部皮下注射(150 mg·kg^-1·d^-1,共2 d)得到.手术结扎方法是通过手术结扎左前降支完成.模型完成后通过全细胞膜片钳技术测定神经元的延迟整流钾通道和钠通道特性改变.结果 对照组、ISO注射组和手术结扎组的SCG神经元延迟整流钾通道的峰值电流密度分别为80.80±22.80 pA/pF、99.89±21.50 pA/pF和115.33±16.00 pA/pF.三者的延迟整流钾通道激活曲线差异无统计学意义.对照组、ISO注射组和手术结扎组的SCG神经元钠通道的峰值电流密度分别为-102.71±9.55 pA/pF、-144.79±24.34 pA/pF和-125.66±32.34 pA/pF.ISO注射和手术结扎均使钠通道激活曲线和失活后恢复曲线显著左移,但对失活曲线没有显著性影响.结论 通过ISO注射和手术结扎方式得到的两种MI模型,对SCG神经元通道特性的影响是一致的.因此,在进行MI后交感神经节神经元通道特性的研究时两种方法都是可靠的选择.Objective To compare the characteristics of the superior cervical ganglion (SCG) neuron channels in rabbit myocardial infarction (MI) models established by drug injection and surgical ligation,respectively. Methods The MI model established by drug injection was obtained by using abdominal subcutaneous injection of Isoproterenol (ISO,150 mg · kg^-1 · d^-1,for 2 days). The MI model established by surgical ligation was obtained by ligation of the left anterior descending artery. After establishment of the models,the characteristics of the delayed rectifier potassium channel and sodium channel of the neurons were measured by whole-cell patch-clamp technique. Results The maximal peak current densities of delayed rectifier potassium channels of SCG neurons in the control group,ISO injection group and ligation group were 80.80 ± 22.80 pA/pF,99.89 ± 21.50 pA/pF and 115.33 ± 16.00 pA/PF, respectively. There was no significant difference in the delayed rectifier potassium channel activation curve among the three groups. The maximal peak current densities of delayed rectifier sodium channels of SCG neurons in the control group,ISO injection group and surgical ligation group were-102.71 ± 9.55 pA/pF,-144.79±24.34 pA/pF and-125.66±32.34 pA/pF,respectively. The activation curve and recovery curve after inactivation of sodium channel in ISO injection group and ligation group were shifted toward left,whereas there was no significant effect on the inactivation curve. Conclusion ISO injection and surgical ligationon show the same effects on the characteristics of SCG neurons channels in the MI models. Therefore,both methods are viable in the research of the characteristics of neuron channels of sympathetic ganglion after the establishment of MI.
