摘 要:目的通过观察急性白血病(AL)患者外周血浆细胞样树突状细胞(p DCs)的功能及Toll样受体7(TLR7)及TLR9mRNA的表达变化,探讨p DCs功能缺陷的可能原因。方法免疫磁珠分选11例初诊未治急性淋巴细胞白血病(ALL)、13例急性髓细胞白血病(AML)和15例健康对照(正常对照组)外周血p DCs,用Cp G ODN 2216刺激外周血p DCs,收集细胞培养上清液,ELISA检测上清液IFN-α、TNF-α、IL-6水平;real-time PCR检测p DCs TLR7 mRNA、TLR9 mRNA表达。结果 AML组和ALL组p DCs产生的IFN-α、TNF-α、IL-6水平均明显低于正常对照组(P<0.05),p DCs功能明显下降;AML组和ALL组p DCs内TLR7 mRNA、TLR9 mRNA亦明显低于正常对照组(P<0.05)。结论急性白血病源p DCs内TLR7 mRNA、TLR9mRNA表达减少,可能进一步抑制p DCs功能,是AL发病机制之一。Objective To explore the causes of plasmacytoid dendritic cells( p DCs) dysfunction by observing the function of p DCs and Toll-like receptor 7( TLR7) mRNA and TLR9 mRNA expression in patients with acute leukemia( AL). Methods The peripheral blood p DCs from 11 patients with untreated acute lymphocytic leukemia( ALL),13 patients with acute myeloid leukemia( AML) and 15 healthy controls( normal control group) were sorted by immune magnetic beads. After the p DCs were cultured with Cp G ODN 2216,the supernatants were collected to detect the levels of IFN-α,TNF-α and IL-6 in supernatants by ELISA and the expression of TLR7 mRNA and TLR9 mRNA in p DCs by real-time PCR. Results The levels of IFN-α,TNF-α and IL-6 produced by p DCs were significantly lower in patients with AML and ALL than those of normal controls( P〈0. 05),and the p DCs function significantly decreased. Compared with controls,the mRNA expression of TLR7 and TLR9 in p DCs was significantly decreased in AML and ALL patients( P〈0. 05). Conclusion The expression of TLR7 mRNA and TLR9 mRNA in p DCs of AL decrease,which further inhibits the function of p DCs and promote the pathogenesis of AL.
