摘 要:盾叶薯蓣是中国特有的甾体激素类药源植物,但关于其种质资源遗传多样性和分子标记辅助育种的研究相对较少。本研究在盾叶薯蓣转录组分析的基础上,进一步挖掘其SNP和SSR分子标记。研究发现,盾叶薯蓣转录组的37 115条unigene中含有124 692个SNP位点,平均每条unigene含3.36个SNP位点。用软件MISA对盾叶薯蓣转录组进行搜索发现13 514个SSR位点分布于11 615条unigene序列中,SSR发生频率为13.66%,平均每7.18 kb含1个SSR位点。共筛选设计出4 329对SSR引物。本研究不仅丰富了盾叶薯蓣的分子标记,为其遗传资源评价、种质资源鉴定与改良提供帮助,而且对盾叶薯蓣功能基因资源的开发利用及其比较基因组学研究具有重要的价值。Dioscorea zingiberensis C. H. Wright is the unique plant source for steroid hormone medicines in our country. But little researches had been carried out on genetic diversity of germplasm resources and breeding assisted by molecular marker. Based on transcriptome of D. zingiberensis, SNP and SSR markers were mined. A total of 124 692 SNP sites were found from 37 115 unigenes, and 3.36 SNP sites were found in each unigene on average. 13 514 SSR sites were found from 11 615 unigenes searched by Micro SAtellite(MISA) software. On average, SSRs occurred every 7.18 kb in length with an occurrence frequency of 13.66%. A total of 4 329 primer pairs were designed for marker development. This study not only enriches the molecular markers of D. zingiberensis and lay a foundation for the evaluation of genetic resources, germplasm identification and improvement, but also have important value for utilization of genetic resources and comparative genomics research in D. zingiberensis.
