摘 要:目的:探讨孤儿受体ERRα对前列腺癌细胞能量代谢转化的影响。方法:通过慢病毒介导的FLAG构建ERRα表达稳定上调的LNCa P前列腺癌细胞模型,Western Blotting法检测其表达量,检测常氧和低氧条件下培养的LNCa P和LNCa P-ERRα细胞相关酶活性(乳酸脱氢酶、己糖激酶、丙酮酸激酶)及ADP含量。结果:过表达ERRα的LNCa P细胞的乳酸脱氢酶、己糖激酶、丙酮酸激酶(低氧)活性和ADP的量均明显高于LNCa P细胞(P<0.05)。结论:在前列腺癌细胞中上调ERRα的表达水平能促进其能量代谢转化。Objective:To investigate the effect of orphan receptor ERRα on energy metabolism and transformation of prostate cancer cells. Methods:LNCaP prostate cancer cell model with stable up-regulated expression of ERRα was established by lentivirus-mediated FLAG. The expression level was determined by Western blotting. Cultured under normoxia and hypoxia conditions,the LNCaP and LNCaP-ERRαcells-associated enzyme activities (lactate dehydrogenase,hexokinase and pyruvate kinase) and ADP contents were measured. Results:The activities of lactate dehydrogenase,hexokinase and pyruvate kinase ( hypoxia) and ADP contents in ERRα over-expressing LNCaP cells were significantly higher than those in LNCaP cells ( P〈0.05) . Conclusion:Up-regulating the expression level of ERRα in prostate cancer cells can promote its energy metabolism and transformation.
