摘 要:目的探讨KISS-1基因及其受体GPR54的表达与先兆子痫发生的相关性。方法以实时定量RT-PCR检测先兆子痫患者胎盘滋养层细胞KISS-1基因及其受体GPR54 m RNA表达,以免疫组化和western-blot检测Kisspeptin蛋白表达。计量资料比较采用t检验,P<0.05为差异有统计学意义。结果胎盘滋养层组织均表达KISS-1基因和其受体GPR54;先兆子痫患者胎盘KISS-1/GPR54免疫组化染色强度、m RNA及蛋白表达量均高于正常胎盘(均P<0.05)。结论 KISS-1/GPR54的异常表达可能是先兆子痫的发病机制。Objective To investigate the correlation between the occurrence of preeclampsia and the expression of KISS-1 gene(melanoma metastasis suppressor gene) and its receptor GPR54(G protein coupled receptor).Methods The expression of KISS-1 gene and its receptor GPR54 m RNA in placental trophoblast cells of preeclampsia patients were detected by real-time quantitative RT-PCR, and the expression of Kisspeptin protein was detected by immunohistochemistry and western blot. Measurement data uses t test, P0.05 is considered statistically significant.Results KISS-1 and its receptor GPR54 expressed in the placental trophoblast tissues.The immunohistochemical staining intensity of KISS-1/GPR54 and the expression of m RNA and protein in preeclampsia placenta were all significantly higher than those in normal placenta(all P0.05). Conclusion The abnormal expression of Kisspeptin/GPR54 may be involved in the pathogenesis of preeclampsia.
