摘  要:目的:探讨16 Hz,150 dB次声作用1、3、5 d对大鼠海马caspase-1、caspase-3表达的影响。方法:将成年雄性SD大鼠置于高强度次声仓内,进行16 Hz,150 dB强度的次声照射,每天2 h。40只大鼠随机分为四组,对照组、次声组(分别为次声1 d组、3 d组和5 d组)。次声照射结束后,立即取材。采用免疫荧光法观察大鼠海马caspase-3的表达变化,采用Western Blot方法检测caspase-1、caspase-3的表达量。结果:免疫荧光染色结果显示:与正常组相比,次声5 d组的大鼠海马caspase-3的表达增多(P<0.05);Western Blot结果显示:与正常组相比,次声5 d组的大鼠海马caspase-1的表达增多、caspase-3的表达减少、活化形式的caspase-3表达增多(P<0.05);次声5 d组大鼠海马caspase-1、caspase-3的表达均比次声1 d组和次声3 d组要高(P<0.05)。结论:16 Hz,150 dB的次声随着对大鼠作用的时间延长,其海马caspase-1和caspase-3的表达逐渐增多,引起海马组织的焦亡和凋亡也增多。Objective: To investigate the effects of 16 Hz, 150 dB infrasound on the expression of caspase-1 and caspase-3 in rat hippocampus at 1, 3, 5 d. Methods: Adult male SD rats were placed in a high-intensity infrasound chamber and subjected to infrasound at 16 Hz and 150 dB intensity for 2 hours per day. Fourty male Sprague-Dawley rats were randomly divided into four groups: control group, irffrasound group (after exposure for 1, 3, 5 d to 16 Hz 150 dB infrasound). The expression change of caspase-3 in the hippocampus of rats was observed by immunofluorescence histo- chemistry. The expression level of caspase-1 and caspase-3 was detected by Western Blot. Results: Immunofluorescence histochemical results showed that the expression of caspase-3 in the hippocampus of the infrasound group at the fifth day was significantly higher than that of the normal group (P 〈 0.05). Western Blot results showed that the expression levelof caspase-1 and cleaved caspase-3 in the hippocampus of the infrasound group was significantly higher than that in the in- frasound group (P 〈0.05). Conclusion: The infrasound of 16 Hz and 150 dB has the greatest effect on caspase-1 and caspase-3 in hippocampus of rats and the apoptosis ceils were significantly increased in the infrasound group.
