摘  要:目的探讨胡黄连苷Ⅱ在大鼠睾丸缺血再灌注损伤过程中的保护作用。方法选取成年雄性SD大鼠45只随机分为3组,每组15只:睾丸缺血再灌注+胡黄连苷Ⅱ组(睾丸IRI+胡黄连苷Ⅱ组);睾丸缺血再灌注组(睾丸IRI组);对照组。睾丸IRI组和睾丸IRI+胡黄连苷Ⅱ组大鼠分别建立睾丸扭转复位模型,睾丸IRI+胡黄连苷Ⅱ组在再灌注同时腹腔注射胡黄连苷Ⅱ(10 mg/kg)。HE染色观察睾丸组织病理改变;测定丙二醛(MDA)、超氧化物岐化酶(SOD)含量;采用原位缺口末端标记检测细胞凋亡指数;免疫组化检测睾丸组织中的Bax和Caspase-3的表达情况;RT-PCR检测TNF-α和IL-1βmRNA表达水平。结果睾丸IRI组可见生精上皮排列紊乱,生精细胞广泛脱落等表现;3组的凋亡指数分别为(4.28±0.36)%、(18.93±1.27)%、(9.53±1.02)%,SOD活性分别为(0.83±0.05)、(0.28±0.03)、(0.54±0.03)U/mg蛋白,MDA含量分别为(1.92±0.26)、(5.23±0.78)、(2.97±0.35)nmol/g蛋白,各组上述指标相互比较均有统计学意义(P<0.05);睾丸IRI组中Bax及Caspase-3表达明显高于对照组,睾丸IRI+胡黄连苷Ⅱ组明显改善IRI引起的表达变化;与对照组相比,睾丸IRI组中TNF-α和IL-1βmRNA表达水平明显增高。和睾丸IRI组比较,睾丸IRI+胡黄连苷Ⅱ组中TNF-α和IL-1βmRNA表达水平明显减低,各组上述指标相互比较均有统计学意义(P<0.05)。结论胡黄连苷Ⅱ可减轻氧化应激水平,发挥抗炎及抗凋亡作用,从而在睾丸组织在缺血再灌注损伤过程中发挥保护作用。[Objective] To investigate the protective effects of picroside Ⅱ on testicular ischemia reperfusion injury in rats.[Methods]Forty-five adult male SD rats were randomly divided into 3 groups,15 rats in each group: testicular ischemia reperfusion injury + picroside Ⅱ group(testicular IRI+picroside Ⅱ);testicular ischemia-reperfusion injury group(testicular IRI);control group. The rats of testicular IRI and testicular IRI+picroside Ⅱ groups respectively established testicular torsion reduction model,and picroside Ⅱ(10 mg/kg) was injected intraperitoneally followed reperfusion in testicular IRI+picroside Ⅱ group rats.HE staining was used to observe the pathological changes of testis;the contents of malondialdehyde(MDA) and superoxide dismutase(SOD) were detected;cell apoptosis index was detected by d UTP nick end labeling;the expression of Bax and Caspase-3 in testis tissue was detected by immunohistochemistry;and the mRNA expression levels of TNF-α 和 IL-1βwere measured by RT-PCR. [Results] The testicular IRI group showed disordered arrangement of spermatogenic epithelium and detachment of spermatogenic cells. Apoptotic indexes in above three groups were(4.28±0.36)%,(18.93±1.27)% and(9.53±1.02)%,respectively. The viability of SOD were(0.83 ±0.05),(0.28 ±0.03) and(0.54 ±0.03) U/mg protein. MDA contents were(1.92±0.26),(5.23 ±0.78) and(2.97 ±0.35) nmol/g protein. The differences of above indexes were statistically significant(P 0.05).Compared with control group,the expression levels of Bax and Caspase-3 in testicular IRI group were obviously increased,and the testicular IRI+picroside Ⅱ group improved the expression changes induced by IRI. Compared with control group,the mRNA expression levels of TNF-α and IL-1βwere significantly increased.
