肌肽对H_2O_2、Aβ_(25-35)诱发PC12细胞损伤的保护作用

摘　　要：目的　研究肌肽对H2 O2 和 β淀粉样蛋白片段 (Aβ2 5- 3 5)诱发PC12细胞损伤的保护作用。方法　将培养的PC12细胞分为 3组 :正常对照组 ;损伤组 ,分别加入H2 O2 和Aβ2 5- 3 5;保护组 ,在用H2 O2 、Aβ2 5- 3 5处理前 30min加入肌肽。用LDH法、MTT法观察肌肽的神经保护作用。结果　保护组LDH活性显著少于损伤组 (P <0 .0 1) ;MTT测定结果显著高于损伤组 (P <0 .0 1)。结论　肌肽对H2 O2 和Aβ2 5- 3Objective To study the protective effect of carnosine against toxic effect of hydrogen peroxide(H 2O 2) and β-amyloid(25-35)(Aβ 25-35) on PC12 cells.Methods Culturing PC12 cells were divided into 3 groups:control group,damage group in which cells were mixed with H 2O 2 and Aβ 25-35 respectively,protecting group in which carnosine were added 30 minutes before.LDH and MTT experiment were used.Results LDH in protecting group was significantly lower than that of damage group(P<0.01).OD value obtained by MTT in protecting group was obviously higher than that of damage group(P<0.01).Conclusion Carnosine can protect PC12 cells from toxic effects of H 2O 2 and Aβ 25-35.
