摘 要:目的研究上皮性卵巢癌患者血浆中micro RNA-30a-5p与卵巢癌分级、分期的关系,探讨其在卵巢癌中的诊断价值。方法选取111例上皮性卵巢癌患者,患者病理分级为低级上皮性卵巢癌52例,高级上皮性卵巢癌59例,根据国际妇产科联盟(International Federation of Gynecology and Obstetrics,FIGO)分期标准:Ⅰ期24例,Ⅱ期27例,Ⅲ期32例,Ⅳ期28例;同期健康女性体检者20例,采用实时荧光定量PCR检测血浆中micro RNA-30a-5p的表达,分析micro RNA-30a-5p表达与上皮性卵巢癌分级及分期的关系。结果与正常对照组比较,上皮性卵巢癌患者血浆中micro RNA-30a-5p明显下降,差异有统计学意义(P<0.05),高级上皮性卵巢癌患者血浆中micro RNA-30a-5p表达量明显低于低级(P<0.05);随着肿瘤分期恶性度的升高,micro RNA-30a-5p相对表达量下降,但Ⅰ期与Ⅱ期结果差异无统计学意义(P>0.05),其余各组差异均有统计学意义(P<0.05)。结论 micro RNA-30a-5p的表达与上皮性卵巢癌发生、发展以及肿瘤的恶性度有关,为肿瘤的靶向治疗提供了依据。Objective To investigate the relationship between plasma micro RNA-30 a-5 p level and grading and staging of ovarian cancer, and to explore the diagnostic significance for epithelial ovarian cancer.Methods A total of 111 cases of patients with epithelial ovarian cancer were recruited, 52 cases of patients with pathological grading for low-grade epithelial ovarian cancer, 59 case for advanced epithelial ovarian cancer. The patients were divided into 4 groups:stageⅠ(24 cases), Ⅱ(27 cases), Ⅲ(32 cases) and Ⅳ(28 cases),according to the International Federation of Gynecology and Obstetrics(FIGO). 20 healthy female subjects were served as controls. The expression of micro RNA-30 a-5 p in plasma was detected by quantitative real-time PCR, and the relationship between the expression of micro RNA-30 a-5 p and the grading and staging of epithelial ovarian cancer was analyzed. Results Compared with the normal control group, micro RNA-30 a-5 p in the plasma of patients with epithelial ovarian cancer was significantly decreased(P〈0.05). The expression of micro RNA-30 a-5 p in patients with advanced epithelial ovarian cancer was significantly lower than that in low-grade ovarian cancer(P〈0.05). With the malignant degree of tumor staging increased, the relative expression of micro RNA-30 a-5 p decreased among the other groups(P〈0.05), except stageⅠandⅡ(P〉0.05).Conclusion The expression of micro RNA-30 a-5 p is related to the occurrence, development and malignant degree of epithelial ovarian cancer, which may served as a novel diagnostic marker, and be used as a therapeutic target for the treatment of epithelial ovarian cancer.
