六味地黄方对诱发食管腺癌大鼠模型预防作用研究

摘　　要：目的:探索六味地黄方对Barrett食管进展为食管腺癌的预防作用。方法:40只SD大鼠行食管空肠吻合术建立Barrett食管诱发腺癌动物模型,随机分为六味地黄方组和对照组,分别予六味地黄颗粒溶液、生理盐水灌胃,并加用右旋糖酐铁促癌模拟癌变病程,28周后统一处死,镜下观察食管异型增生、Barrett食管、腺癌的发病率。结果:两组均可见严重食管炎、异型增生、Barrett食管和腺癌,六味地黄颗粒组腺癌发生率为9.09%(1/11),明显低于对照组的36.36%(4/11),但差异并无统计学意义(P>0.05)。结论:六味地黄方有降低Barrett食管癌变的趋势,可能对阻断Barrett食管进展为腺癌有一定的作用,有待进一步研究。Objective ： To discuss the preventive effect of Liuwei Dihuang Decoction in an animal model of Barrett＇s esophagus. Methods ： 40 SD rats that underwent an esophagojejunostomy with gastric preservation were divided into two groups. One group was perfused with Liuwei Dihuang Particles liquid （ traditional Chinese medicine group, TCMG ） everyday, and the other was given same amount of saline （ control group, TG ）. All of the rats were received a peritoneal injection of iron dextran every two weeks. The animals were killed at 28 weeks after gastric perfusion. Pathological evaluation was performed with HE staining. Results ： Severe esophagitis, irregular hyperplasia, Barrett＇s esophagus and esophageal adenocarcinoma can be observed in both two groups. The incidence rate of esophagus adenocarcinoma in TCMG was 9.09% （ 1/11 ）, obviously lower than that of TG, which was 36.36% （ 4/11 ）, but there was no significant difference between the two groups （ P〈0.05 ）. Conclusion： Traditional Chinese medicine Liuwei Dihuang Decoction showed the probability to reduce the chance of esophageal adenocarcinoma and it may be effective in preventing the development of esophageal adenoearcinoma from Barrett＇s esophagus, further research is needed.
