摘 要:目的 :研究苦参(Sophora flavescenes Ait)总生物碱及其主要生物碱的抑菌效果,探究苦参开发消洗剂的可行性。方法 :采用大孔吸附树脂柱色谱、硅胶柱色谱等技术手段对苦参干燥根进行分离纯化并通过波谱数据对化合物进行结构鉴定。采用药物二倍稀释法与菌落计数相结合法考察苦参总生物碱及其主要生物碱对金黄色葡萄球菌、大肠埃希杆菌、铜绿假单胞菌、乙型溶血型链球菌和白色念珠菌的抑菌作用,并测定各成分用药时的抑菌浓度范围。结果 :从苦参中分离了16个单体化合物,包括9个生物碱类成分和7个黄酮类成分。药敏结果显示,5个样品对所选菌株均具有一定抑菌活性;苦参总生物碱抑菌作用强于各单体生物碱;各单体对金葡球菌的抑菌效果强于其他4种菌;苦参碱和槐果碱对金黄色葡萄球菌的抑菌作用均强于自身氧化物。结论 :苦参中总生物碱及其主要生物碱成分存在明显的抑菌作用,各单体间抑菌活性各有不同,且对各菌种的抑菌作用也不甚相同,可为苦参开发消洗剂的可行性奠定基础。Objective :To study the chemical composition and antimicrobial activity of Radix Sophorae flavescentis,and to use it for the add-on development. Methods :The root of Radix Sophorae flavescentis were isolated and purified from by macroporous resin column chromatography,silica gel column chromatography,Sephadex LH-20 column chromatography,reverse phase C_(18) open column chromatography and preparation of HPLC and other technical means. The drug sensitivity of the total alkaloids and their main alkaloids were investigated by the method of two times dilution and colony counting. Results :Sixteen compounds were isolated from Sophora flavescens,including 9 alkaloids and 7 flavonoids. The result shows that the total alkaloids of Sophora flavescens and its main alkaloid has certain antibacterial activity against Staphylococcus aureus,Escherichia coli,Pseudomonasaeruginosa,Beta hemolytic streptococcus and Candida albicans. The inhibitory effect of total alkaloids of the Radix Sophora flavescens was stronger than the individual compounds,and the antibacterial effect of Staphylococcus aureus and Candida albicans was better. The inhibitory effect of the individual compounds to Staphylococcus aureus was stronger than the other four strains. Inhibitory effects of matrine and sophocarpine to Staphylococcus aureus was stronger than its oxide. Conclusion :The total alkaloids and their main alkaloids of Radix Sophorae flavescentis have obvious antimicrobial effect,and the antibacterial activity of each monomer was different,which could be used as the basis for the development of two times.
