摘 要:目的观察口腔鳞状细胞癌(OSCC)组织中转录因子Twist及上皮细胞钙黏蛋白(E-cadherin)的表达变化,并探讨其意义。方法采用免疫组化SP法检测65例OSCC组织、45例正常口腔黏膜组织中Twist和E-cadherin蛋白表达,分析Twist、E-cadherin表达与OSCC患者临床病理特征的关系,并采用Spearman等级相关分析法分析OSCC组织中Twist和E-cadherin表达的关系。结果 Twist在OSCC组织和正常口腔黏膜组织中的阳性表达率分别为72.31%(47/65)、11.11%(5/45),差异有统计学意义(P<0.05);E-cadherin在OSCC组织和正常口腔黏膜组织中的阳性表达率分别为23.08%(15/65)、86.67%(39/45),差异有统计学意义(P<0.05)。OSCC组织中,Twist和E-cadherin异常表达与患者淋巴结转移和肿瘤临床分期有关(P均<0.05),与患者性别、年龄、组织分化程度、生长类型无关(P均>0.05)。OSCC组织中,Twist和E-cadherin表达呈负相关(r=-0.412,P<0.05)。结论 OSCC癌组织中转录因子Twist表达上调,E-cadherin表达下调;二者在OSCC发生和恶性进展中发挥重要作用。Objective To explore the expression changes of the transcription factor Twist and the E-cadherin in the oral squamous cell carcinoma( OSCC) tissues and to investigate the clinical significance. Methods The immunohistochemical SP method was used to detect the expression levels of the Twist and E-cadherin in 65 cases of OSCC tissues and45 cases of oral normal mucosa tissues. In addition,the relationship between the expression of Twist/E-cadherin and the clinicpathologic characteristics was analyzed,and the Spearman was used to explore the correlation between the Twist and E-cadherin in OSCC tissues. Results The positive expression rates of Twist in the OSCC tissues and oral normal mucosa tissues were 72. 31%( 47/65) and 11. 11%( 5/45),respectively( P〈0. 05). The positive expression rates of E-cadherin in the OSCC tissues and oral normal mucosa tissues were 23. 08%( 19/65) and 86. 67%( 39/45),respectively( P〈0. 05). The Twist and E-cadherin expression levels in the OSCC tissues were related with the lymph node metastasis and the clinical stages( both P〈0. 05),but were not related with the gender,age,differentiation degree and the type( all P〉0. 05). The expression of Twist and E-cadherin was negatively correlated in the OSCC tissues( r =-0. 412,P〈0. 05).Conclusions The transcription factor Twist is up-regulated and E-cadherin is down-regulated,both of them play a critical role in the occurrence and malignant progression of OSCC.
