摘  要:目的筛选可作为胃癌诊断生物标志物的微小核糖核酸(miRNA)。方法提取100例胃癌患者和60例健康体检者的血清RNA,采用定量PCR芯片分析和荧光定量PCR检测胃癌患者血清miRNA水平的异常,采用ROC曲线分析候选miRNA对胃癌的诊断效能。结果与健康体检者相比,在胃癌患者中鉴定得到5个水平上调和4个水平下调的miRNA;ROC曲线分析验证,这9个miRNA共同诊断胃癌的AUC为0.934[95%CI(0.877~0.992)](P<0.01)。结论检测多种在胃癌患者中水平异常的miRNA组合能够作为胃癌诊断的潜在指标。Objective To screen serum microRNA(miRNA)as the potential biomarkers for the diagnosis of gastric cancer.Methods Plasma RNA was extracted from 100 patients with gastric cancer and 60 healthy people.The abnormity in serum level of miRNA in the patients with gastric cancer was detected by quantitative-PCR microarray analysis and fluorescence quantitative-PCR.The diagnostic efficacy of candidate miRNA in the diagnosis of gastric cancer was evaluated by ROC curve analysis.Results Compared with healthy people,5 upregulated and 4 downregulated miRNAs were identified in the patients with gastric cancer.ROC curve analysis verified that the AUC of combination of these 9 miRNA for the diagnosis of gastric cancer was 0.934[95% CI(0.877-0.992)](P〈0.01).Conclusion Detecting abnormal levels of multiple miRNA could be used as a potential biomarker for the diagnosis of gastric cancer.
