摘  要:利用不同剂量髓鞘少突胶质细胞糖蛋白(Myelin oligodendrocytes glycoprotein,MOG34-56)免疫诱导制备食蟹猴(Macaca fascicularis)实验性自身免疫性脑脊髓炎模型(Experimental autoimmune encephalomyelitis,EAE)。选择4岁左右雌性食蟹猴,分高、低2个剂量组,利用合成的MOG34-56与完全弗氏佐剂(Complete Freund's adjuvant,CFA)混合制备成乳剂,经腋窝和腹股沟皮下多点免疫注射的方法,制备EAE食蟹猴模型,从行为学、核磁共振成像(Magnetic Resonance Imaging,MRI))和病理学等多方面进行评估。结果显示利用不同剂量MOG34-56在完全弗氏佐剂条件下通过皮下多点免疫方法能成功诱导出不同病程的EAE食蟹猴模型,且诱导发病率达100%。表现为行为学异常,体重下降,神经系统显示功能障碍,MRI影像及解剖病理观察均有明显病变,且高剂量诱导组症状较低剂量组严重。To establish EAE cynomolugus monkeys(Macaca fascicularis) model by immunizing myelin protein,4 years old female cynololgus monkeys were selected immunized by the MOG34-56 polypeptide which were divided into high and low dose groups mixed with complete freund's adjuvant by subcutaneous multipoint immunization in the armpit and inguinal region to establish the EAE model. The behavior,MRI imaging and pathology were evaluated. The results showed that MOG34-56 in CFA could successfully induce different course of EAE cynomolgus monkey model by subcutaneous multipoint immune approaches,and the incidence was 100%.Abnormal behavior,weight loss,nervous system dysfunction,obvious lesions in MRI images and anatomical,pathological observation were found in the model monkeys. The symptom of high dose induction group was worse than that of low dose group.
